Introduction. Traumatic ulcers are common lesions in Indonesia, arising from inflammation in the oral cavity. Petai Cina (Leucaena leucocephala), a tropical plant native to Indonesia, contains flavonoids that act as anti-inflammatory agents by reducing levels of pro-inflammatory cytokines, including interleukin-6 (IL-6). Purpose of this study was to determine the effectiveness of petai cina leaf extract with concentrations of 15%, 30%, and 45% in reducing clinical inflammatory symptoms in thermal traumatic ulcers, to analyze the effect of petai cina leaf extract on interleukin-6 (IL-6) levels, and to analyze the relationship between IL-6 levels and clinical symptoms of inflammation. Method This study is a purely experimental in vivo study, with a post-test only control group design. This study used 30 male Wistar rats divided into groups with traumatic ulcers without treatment, povidone iodine treatment, and treatment with 15%, 30%, and 45% Chinese petai leaf extract gel for 7 days. IL-6 levels were examined using the ELISA technique from Wistar rat traumatic ulcer tissue samples. The results of the study based on descriptive tests showed significant differences in the mean values of clinical symptoms of ulcer diameter length, rubor, tumor, and dolor, as well as different IL-6 levels in each treatment group (p<0.05). The Kruskal-Wallis test showed a significant relationship between clinical symptoms of ulcers and IL-6 levels with opposite gel concentrations, as well as a significant relationship between clinical symptoms of ulcers and IL-6 levels that were unidirectional. The conclusion of this study is that there is an effect of petai cina leaf extract gel concentrations of 15%, 30%, and 45% on clinical symptoms of inflammation and IL-6 levels in Wistar rats, a model of traumatic ulcers. There was a significant, one-way relationship between IL-6 levels and clinical symptoms of traumatic ulcers. The results of the treatment group that most closely matched the positive control group with povidone iodine were those with 45% petai cina leaf extract gel.en_US
