dc.description.abstractFixed orthodontic appliances facilitate plaque and biofilm formation, serving as a reservoir for pathogenic bacteria and increasing the risk of caries, gingivitis, and periodontal disease. Due to the limitations of synthetic antibacterials, there is a growing interest in natural alternatives like green betel leaf (Piper betle Linn), which possesses antibacterial properties capable of inhibiting bacterial growth on orthodontic devices. This study aims to assess the antibacterial effectiveness of a green betel leaf-based mouthwash against Staphylococcus aureus ATCC® 29213™ on nickel titanium (Ni-Ti) orthodontic wires. The study comprised five research groups: green betel leaf (Piper betle Linn) mouthwash at concentrations of 3%, 4%, and 5%; a positive control group (0.2% chlorhexidine); and a negative control group (distilled water). The research was initiated by preparing the green betel leaf mouthwashes at the 3%, 4%, and 5% concentrations. Subsequently, antibacterial testing was performed on the Staphylococcus aureus-contaminated wires using the Total Plate Count (TPC) method. Data were analyzed using One-Way ANOVA. The mean of Staphylococcus aureus colony counts were: 637.33±63.78 CFU/ml for the 3% green betel leaf mouthwash group; 559.83±19.395 CFU/ml for the 4% group; 508.67±17.108 CFU/ml for the 5% group; 1.00±0.632 CFU/ml for the 0.2% Chlorhexidine positive control; and 1789.50±120.394 CFU/ml for the distilled water negative control. The results of the statistical analysis indicated a significant difference among all treatment groups (p<0.05). Based on these findings, it is concluded that the green betel leaf (Piper betle Linn) mouthwash demonstrates antibacterial effectiveness against Staphylococcus aureus.en_US
