    Aktivitas Antibakteri Krim Antijerawat yang Mengandung Ekstrak Daun Sirih Hijau (Piper betle L.) Terhadap Propionibacterium Acne

    Abstract
    Background: Acne is a skin disease caused by chronic inflamation of pilosebacea follicles. One of bacteria that causes of acne is the Propionibacterium acne. The leaf of green betel plant (Piper betle L.) contains saponin, tannin, flavonoids, polyphenols, and essential oil as an antibactetial. In this study the green betel leaf extract is formulated in cream dosage form because the cream is easy to apply, good absorbing capability and provide coolness to the skin . Purpose: To determine the antibacterial activity of green betel leaf cream and extract against Propionibacterium acne. Method: Antibacterial activity properties assessment test of green betel leaf cream and extract was done using the disfussion method by paper discs against Propionibacterium acne. The concentrations of the betel leaf extract cream used were (F1) 0%, (F2) 6%, (F3) 7.5, (F4) 9% and (F5) 10% respectively. The physical quality evaluations of the anti acne cream green betel leaf were as such : homogenity test, pH test, physical stability of the preparation, emulsion type test and irritation test by usage test method to volunteers. Result: The results of the antibacterial activity test of the green betel leaf extract against Propionibacterium acne were as such: the minimum inhibitory concentration (MIC) was obtained at the concentration of 15 mg/ml with average inhibition diameter of 6.43 mm and the most active of inhibitory concentration was obtained at the concentration of 100 mg/with average inhibition diameter of 19 mm. The green betel leaf extract was able to be formulated as anti acne cream preparation and had met the requirements of physical quality test. The antibacterial activity of green betel leaf anti acne cream with the concentrations of 6%, 7.5%, 9% and 10% had the average inhibition diameters against Propionibacterium acne of 14.96; 16.26; 18.13; and 19.16 mm respectively. Conclusion: Anti acne cream of green betel leaf extract with 10% concentration had the greatest inhibition potency against Propionibacterium acne bacteria 19.16 mm.
     
    Latar Belakang: Jerawat adalah penyakit kulit akibat peradangan dari folikel pilosebasea. Salah satu bakteri yang menyebabkan jerawat adalah Propionibacterium acne. Daun sirih hijau mengandung saponin, tanin, flavonoid, polifenol dan minyak atsiri sebagai antibakteri. Pada penelitian ini ekstrak daun sirih hijau diformulasikan dalam bentuk sediaan krim karena krim mudah diaplikasikan, daya menyerap yang baik dan memberikan rasa dingin pada kulit. Tujuan: Untuk mengetahui aktivitas antibakteri dari ekstrak dan krim daun sirih hijau terhadap bakteri Propionibacterium acne. Metode: Aktivitas antibakteri dilakukan terhadap ekstrak dan krim daun sirih hijau dengan metode difusi agar menggunakan cakram kertas terhadap bakteri Propionibacterium acne. Konsentrasi krim ekstrak daun sirih hijau yang digunakan (F1) 0%; (F2) 6%; (F3) 7,5%; (F4) 9% dan (F5) 10%. Evaluasi mutu fisik sediaan krim antijerawat yang terdiri dari: uji homogenitas, uji pH, stabilitas fisik sediaan, tipe emulsi dan uji iritasi terhadap sukarelawan dilakukan metode uji pakai. Hasil: Hasil uji aktivitas antibakteri ekstrak daun sirih hijau terhadap Propionibacterium acne diperoleh konsentrasi hambat minimum (KHM) pada konsentrasi ekstrak daun sirih hijau 15 mg/ml dengan rata-rata diameter hambat 6,3 mm dan konsentrasi hambat paling aktif terdapat pada konsentrasi 100 mg/ml dengan rata-rata diameter hambat 19 mm. Ekstrak daun sirih hijau dapat diformulasikan sebagai sediaan krim antijerawat dan memenuhi persyaratan uji mutu fisik sediaan. Aktivitas antibakteri sediaan krim antijerawat ekstrak daun sirih hijau dengan konsentrasi 6%; 7,5%; 9% dan 10% memiliki rata-rata diameter hambat pada bakteri Propionibacterium acne masing-masing adalah 14,96; 16,26; 18,13; dan 19,16 mm. Kesimpulan: Krim antijerawat dengan konsentrasi ekstrak daun sirih hijau 10% mempunyai daya hambat yang paling besar terhadap bakteri Propionibacterium acne adalah 19,16 mm.
