    Pengembangan Metode Penetapan Kadar Bahan Pemanis, Pengawet, dan Kafein Dalam Produk Minuman Ringan Berkarbonasi Menggunakan Kromatografi Cair Kinerja Tinggi

    Abstract
    Soft drink is a popular beverage product and consumed by people. Soft drink contains additional subtances like sweeteners, preservatives, and caffeine. Sweeteners, preservatives, and caffeine can be hazardous to health if they are over consumed. The purpose of this research was to obtained a method to determination simultaneous of acesulfame, saccharin, cyclamate, aspartame, caffeine, benzoate, and sorbate in soft drink using high performance liquid chromatography. This research used high performance liquid chromatography reverse phase two wavelength with LC 20AD detector PDA (Shimadzu), column C18 150 mm x 2.0 mm (Shimadzu), spectrophotometer UV Probe 1800 (Shimadzu), methanol, phosphate buffer, standard material acesulfame-K, saccharin-Na, cyclamate-Na, aspartame, caffeine, benzoate-Na, sorbic acid, and carbonated soft drinks. Carbonated soft drink was collected from a supermarket in Medan City. Amount of samples were four samples and given code A, B, C, and D. Optimation parameter include wavelength, pH of mobile phase, composition of mobile phase, oven’s temperature, and flow rate. Validation parameter include linierity, limit of detection, limit of quantitation, accuracy, precision, and selectivity. The method was obtained then used to determination acesulfame, saccharin, cyclamate, aspartame, caffeine, benzoate, and sorbic acid in carbonated soft drink samples. The result of this research showed that the optimum condition were mobile phase buffer phosphate pH 3.8 : methanol (80 : 20) v/v, flow rate 0.55 ml/minute, oven’s temperature 40 oC, wavelength 200 nm to detect cyclamate, caffeine, aspartame, benzoate, and wavelength 220 nm to detect acesulfame, saccharin, and sorbate. The retention time of acesulfame, saccharin, cyclamate, caffeine, aspartame, benzoate, sorbate were 1.145 minute, 1,571 minute, 2.940 minute, 4.049 minute, 7.533 minute, 8.329 minute, 10.571 minute respectively. The results of validation showed good linierity. Limits of detection of acesulfame, saccharin, cyclamate, aspartame, caffeine, benzoate, and sorbate were 0.013 ppm, 0.032 ppm, 3.341 ppm, 0.026 ppm, 0.017 ppm, 0.018 ppm, 0.048 ppm respectively. Limit of quantitation of acesulfame, saccharin, cyclamate, aspartame, caffeine, benzoate, and sorbate were 0.045 ppm, 0.106 ppm, 11.138 ppm, 0.086 ppm, 0.058 ppm, 0.060 ppm, 0.159 ppm respectively. The result of accuracy test was 95.21% - 99.82% as percent recovery, compatability system test was 1.0% - 1.9% as %RSD, precision test was 1.65 – 2.48% as %RSD, and selectivity showed good results. The result showed that acesulfame, saccharin, cyclamate, caffeine, benzoate, and sorbate in soft drink samples do not exceed maximum limit of usage.
     
    Minuman ringan merupakan produk minuman yang terkenal dan banyak dikonsumsi masyarakat. Minuman ringan mengandung bahan tambahan seperti pemanis, pengawet, dan kafein. Bahan pemanis, pengawet, dan kafein dapat mengganggu kesehatan bila dikonsumsi berlebihan. Tujuan penelitian ini adalah untuk menemukan suatu metode penetapan kadar asesulfam, sakarin, siklamat, aspartam, kafein, benzoat, dan sorbat secara simultan dalam sampel minuman ringan berkarbonasi menggunakan kromatografi cair kinerja tinggi. Penelitian ini mempergunakan kromatografi cair kinerja tinggi fase terbalik dua panjang gelombang dengan instrumen LC 20AD detektor PDA (Shimadzu), kolom C18 150 mm x 2.0 mm (Shimadzu), spektrofotometer UV Probe 1800 (Shimadzu), baku pembanding asesulfam-K, sakarin-Na, siklamat-Na, aspartam, kafein, benzoat-Na, asam sorbat, metanol, dapar fosfat, serta sampel minuman ringan berkarbonasi. Sampel minuman ringan berkarbonasi diperoleh dari sebuah swalayan di Kota Medan. Sampel berjumlah empat buah dan diberi kode A, B, C, dan D. Parameter optimasi meliputi panjang gelombang, pH fase gerak, komposisi fase gerak, suhu oven, dan laju alir. Parameter validasi meliputi linieritas, batas deteksi, batas kuantitasi, akurasi, presisi, dan selektivitas. Metode yang diperoleh kemudian digunakan untuk penetapan kadar asesulfam, sakarin, siklamat, aspartam, kafein, benzoat, dan sorbat dalam sampel minuman ringan berkarbonasi. Hasil penelitian menunjukkan bahwa kondisi optimum pengujian adalah fase gerak dapar fosfat pH 3,8 : metanol (80 : 20, v/v), laju alir 0,55 ml/menit, suhu oven 40 oC, panjang gelombang 200 nm untuk mendeteksi siklamat, kafein, aspartam, benzoat, dan panjang gelombang 220 nm untuk mendeteksi asesulfam, sakarin, dan sorbat. Waktu tambat asesulfam, sakarin, siklamat, kafein, aspartam, benzoat, dan sorbat masing-masing adalah 1,145 menit; 1,571 menit; 2,940 menit; 4,049 menit; 7,533 menit; 8,329 menit; dan 10,572 menit. Hasil validasi menunjukkan linieritas yang baik. Batas deteksi asesulfam, sakarin, siklamat, aspartam, kafein, benzoat, sorbat masing-masing adalah 0,013 ppm; 0,032 ppm; 3,341 ppm; 0,026 ppm; 0,017 ppm; 0,018 ppm; dan 0,048 ppm. Batas kuantitasi asesulfam, sakarin, siklamat, aspartam, kafein, benzoat, dan sorbat masing-masing adalah 0,045 ppm; 0,106 ppm; 11,138 ppm; 0,086 ppm; 0,058 ppm; 0,060 ppm; dan 0,159 ppm. Hasil uji akurasi adalah 95,14% - 99,82% sebagai persen perolehan kembali, uji kesesuaian sistem adalah 1,0% - 1,9% sebagai %RSD, uji presisi adalah 1,65 – 2,48% sebagai %RSD, dan uji selektivitas menunjukkan hasil yang baik. Hasil pengujian menunjukkan bahwa kadar asesulfam, sakarin, siklamat, kafein, benzoat, dan sorbat dalam sampel minuman ringan berkarbonasi tidak melebihi batas maksimal penggunaan.
