dc.description.abstractMakanan Fermentasi disukai karena sifatnya dalam memperpanjang masa simpan, keamanan, fungsi, rasa dan nutrisinya. Beberapa makanan fermentasi telah diuji secara in vitro sebagai penghambat enzim α-glukosidase. Dengke naniura adalah salah satu makanan tradisional Indonesia yang dibuat dengan cara fermentasi. Penelitian ini bertujuan untuk mengetahui identitas bakteri pada dengke naniura dan nilai hambatannya terhadap enzim α-glukosidase. Dengke naniura dibuat dengan merendam ikan mas (Cyprinus carpio) terlebih dahulu dengan asam jungga (Citrus jambhiri) sehingga tercipta suasana asam dan kemudian ditambah bumbu segar untuk meningkatkan rasanya. Isolasi bakteri (BAL) dilakukan dengan pengenceran bertahap dari sampel. Isolat BAL yang terpilih kemudian dikarakterisasi secara morfologi, biokimia dan identifikasinya secara molekular melalui pengurutan DNA. Empat isolat yang didapat (DL-109; DL-107; BL-108 and BL-107) merupakan gram positif dan bentuk batang non motil. Sifat fisiologis dan biokimianya mengkonfirmasi sifat-sifat BAL. Dari keempat isolat didapatkan 2 bakteri yang identik 99% dengan Lactobacillus fermentum dan Aneurinibacillus aneurinilyticus. Pada pengujian terhadap aktivitas penghambatan enzim α-glukosidase isolat BAL DL-109 menunjukkan nilai hambatan yang lebih dominan dibanding akarbose dan ketiga isolat BAL yang lain (BL-108, DL-107, BL-107); dimana pada dosis terendah yaitu 25 μg/mL DL-109 memberikan nilai hambatan sebesar 54,76%. Dari penemuan ini dapat disimpulkan bahwa isolat merupakan BAL dan dapat digunakan untuk menurunkan kadar gula darah melalui penghambatan enzim α-glukosidase.en_US
dc.description.abstractFermented foods were favorable because of its properties in enhancing the shelf life, safety, function, sensory and nutrition. There are many fermented foods tested in vitro as α-glucosidase enzyme inhibitor. Dengke naniura is one of Indonesia’s traditional food made by means of fermentation. This research aim to know the identity of bacteria in dengke naniura and its inhibition on α-glucosidase enzyme. Dengke naniura were made by soaking the common carp (Cyprinus carpio) first with rough lemon (Citrus Jambhiri) so that it create the acid condition and then added fresh spices to enhance the flavor. The isolation of LAB conducted by means of serial dilution of the samples. Selected isolate of LAB was then characterized by its morphology, growth in different temperature and biochemical identification; and its identification molecularly by DNA sequencing. The four isolates (DL-109; DL-107; BL-108 and BL-107) were a gram-positive and non-motile rod. The Physiological and biochemical properties of the isolates confirm that its LAB properties. From the four isolates obtained 2 bacterias which identic 98% with Lactobacillus fermentum dan Aneurinibacillus aneurinilyticus. On the test against α-glucosidase enzyme activity inhibition, isolate DL-109 LAB showed dominan activity compared to Acarbose and three others (BL-108, DL-107, BL-107); while at the lowest dosage of 25 μg/mL DL-109 showed inhibition point as much as 54.76%. These findings concluded that the isolates were LAB by its properties and can be used for lowering blood glucose in term of inhibition of α-glucosidase enzyme.en_US
