Latar Belakang : Nikotin adalah komponen psikoaktif yang mengandung tembakau. Nikotin dimetabolisme di hati menjadi kotinin (COT), dan kemudian menjadi trans-3’-hidroksikotinin (3HC). Biomarker urin dapat memberikan informasi tentang asupan dan metabolism nikotin.
Tujuan : Mengidentifikasi kadar nikotin dan kotinin di dalam urin pada perokok aktif
Metode Penelitian :Sampel konsekutif dan penelitian deskriptif.Seluruh sampel berjenis kelamin laki-laki, perokok atau memiliki riwayat merokok.Peneliti menilai kadar nikotin dan kotinin dengan menggunakan teknik Liquid Chromatography tandem-Mass Spectometry (LC-MS/MS).
Hasil : Penelitian ini meliputi 46 sampel terdiri dari 23 orang perokok aktif dan 23 orang mantan perokok. Diperoleh rerata ± SD nikotin adalah 1276.4 ± 918.5 ng/ml, rerata ± SD kotinin adalah 2210.9 ± 1659.4 ng/ml, rerata ± SD trans-3’-hidroksikotinin adalah 2196.0 ± 2417.3 ng/ml pada perokok aktif.
Kesimpulan : Terdapat peningkatan kadar nikotin dan kotinin di urin pada perokok aktif. Introduction: Nicotine is a psychoactive component contained in tobacco. Nicotine is metabolized in the liver into cotinine (COT), and then into trans-3’-hydroxicotinine (3HC). Urine biomarkers can provide information about the intake and metabolism of nicotine.
Aim: Identified nicotine and cotinine levels in the urine of the active smokers.
Methods: The samples were consecutive, and the study was descriptive. All subjects were male and smokers or had a history of smoking. The nicotine and cotinine levels of subjects were assessed using the Liquid Chromatography tandem-Mass Spectrometry (LCMS/MS).
Results: A total of 46 subjects participated in the study consisting of 23 active smokers and 23 former smokers. The mean ± SD of nicotine obtained was 1276.4 ± 918.5 ng/ml, the mean ± SD of cotinine obtained was 2210.9 ± 1659.4 ng/ml, and the mean ± SD trans-3’-hydroxicotinine was 2196.0 ± 2417.3 ng/ml for active smokers.
Conclusion: There was an increase in the nicotine and cotinine levels in the urine of active smokers.
