Uji Aktivitas Antibakteri Fraksi Ekstrak Daun Titanus (Leea aequata L.) Terhadap Bakteri Staphylococcus aureus, Staphylococcus epidermidis dan Pseudomonas aeruginosa

Tumbuhan titanus (Leea aequata L.) merupakan tumbuhan yang digunakan sebagai obat tradisional.Daun dan rantingnya digunakan sebagai antiseptik dan pengobatan luka.Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri fraksi daun titanus terhadap Staphylococcus aureus, Staphylococcus epidermidis dan Pseudomonas aeruginosa.
Serbuk simplisia diekstraksi dengan maserasi menggunakan pelarut etanol dan ekstrak etanol difraksinasi menggunakan pelarut n-heksana dan etil asetat.Selanjutnya fraksi n-heksana dan etil asetat diskrining dan dilakukan uji aktivitas antibakteri terhadap bakteri Staphylococcus aureus, Staphylococcus epidermidis dan Pseudomonas aeruginosa menggunakan metode difusi agar menggunakan pencadang kertas.
Hasil skrining fitokimia fraksi n-heksana menunjukkan adanya steroida/triterpenoida.Fraksi etil asetat adanya alkaloida, flavonoid, glikosida, saponin, dan tanin dan pada fraksi sisa menunjukkan adanya alkaloida, flavonoid, glikosida dan saponin. Hasil uji aktivitas antibakteri fraksi n-heksana menunjukkan aktivitas antibakteri yang efektif pada konsentrasi 300 mg/ml pada bakteri Staphylococcus aureus dengan diameter daerah hambat 14,20 mm, pada konsentrasi 400 mg/ml pada bakteri Staphylococcus epidermidis dengan diameter daerah hambat 14,23 mm dan pada konsentrasi 500 mg/ml pada bakteri Pseudomonas aeruginosa dengan diameter daerah hambat 14,27 mm. Fraksi etil asetat memiliki efektivitas terbesar sebagai antibakteri yaitu konsentrasi 25 mg/ml pada bakteri Staphylococcus aureus dan Staphylococcus epidermidis dengan diameter daerah hambat 14,93 mm dan 14,47 mm dan konsentrasi 50 mg/ml pada bakteri Pseudomonas aeruginosa dengan diameter daerah hambat 14,83 mm. Fraksi sisa menunjukkan aktivitas antibakteri terlemah yaitu konsentrasi 400 mg/ml efektif terhadap Staphylococcus aureus dan Staphylococcus epidermidis dengan diameter daerah hambat 14,50 mm dan 14,70 mm dan pada konsentrasi 500 mg/ml terhadap Pseudomonas aeruginosadengan diameter daerah hambat 14,07 mm. Leea aequata L. is one of the plants including traditional medicines, its leaf and branches has been used as an antiseptic and wound healing.This study aimsto determinethe antibacterial activity offraction titanus leaves againstStaphylococcus aureus, Staphylococcus epidermidisand Pseudomonas aeruginosa.
Powder of simplex was extracted by maceration using ethanoland ethanol extract continued by fractioning using n-hexane and ethylacetate. Hereafter n-hexane and ethylacetate fraction were screened and tested antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa using agar diffusion with the paper disk.
The pytochemical screening of n-hexane fraction only showed the presence of steroids/triterpenoids. Ethyl acetate fraction resulted there were alkaloids, flavonoid, glycocides, saponins and tannin and the remaining fraction showed the presence of alkaloids, flavonoids, glycocides, and saponin. The result of antibacterial activity test showed that n-hexane fraction effective at 300 mg/ml of concentration against Staphylococcus aureus with inhibition diameter areas were 14.20 mm, at 400 mg/ml of concentration against Staphylococcus epidermidis with inhibition diameter areas were 14.23 mm and at 500 mg/ml of concentration against Pseudomonas aeruginosa with inhibition diameter areas were 14.27 mm. The most effective antibacterial agent was ethyl acetate fraction that obstruct the growth of Staphylococcus aureusand Staphylococcus epidermidisat 25 mg/ml of concentration with inhibition diameter areas were 14.93 mm and 14.47 mm and at 50 mg/ml concentration against Pseudomonas aeruginosa with inhibition diameter areas were 14.83. The remaining fraction showed the poorest effectivity of antibacterial activity at 400 mg/ml concentration against Staphylococcus aureusand Staphylococcus epidermidiswith inhibition diameter areas were 14.50 mm and 14.70 mm and at 500 mg/ml of concentration against Pseudomonas aeruginosa with inhibition diameter areas were 14.07 mm.
