    Senyawa dan Aktivitas Antimikroba Asam Lemak dan Esternya dari Biji Durian (Durio Zibethinus Murr)

    Abstract
    The Research related by extraction fatty acids and synthesis of its ester from durian seed to test antimicrobial activity against bacterial pathogens have been done. Extraction of fatty acids has done by maceration method using n-hexane. Synthesis of fatty acid methyl esters (MEAL) and fatty acid ethyl esters (EEAL) was used H2SO4(p) and NaOCH3 catalysts. The synthesis compounds were analyzed using GC-MS and FT-IR. Compounds of fatty acid (AL), MEAL, and EEAL with concentration variations of 10 %, 20 %, and 30 % were tested for antimicrobial activity against pathogenic bacteria S. Aureus, E. Coli, S. Mutans and C. Albicans by disc diffusion method. The process of maceration of durian seed with n-hexane gives a yield of 3,45 %. Oil seed durian composition of free fatty acids, triglycerides, diglycerides, and monoglycerides. The highest triglyceride molecule is C50-POP 17,5878 %; C54-SLS 14,5857 % and C50-PSP 12,7259 %. The result of the synthesis was analyzed GC-MS, there are six ester compound that is interpretable patterns of fragmentation that is 10-octadecadienoic acid methyl ester, octadecanoic acid methyl ester, arachidonic acid methyl ester, tetradecanoic acid ethyl ester, 9-hexadecadienoic acid ethyl ester, octadecanoic acid ethyl ester, 9-octadecadienoic acid ethyl ester and ethyl ester acidic acid. Antimicrobial activity test showed that the highest activity against C. Albicans was indicated by MEAL 30 % with an antimicrobial index (IAM) = 0,91. While for S. Aureus, E. Coli and S. Mutans the highest activity was shown by AL 30 % with IAM 0,75; 2,93 and 1,38 respectively. Based on the above data, the AL and its ester compound have activity as antimicrobial against S. Aureus, E. Coli, S. Mutans, and C. Albicans.
     
    Penelitian terkait ekstraksi asam lemak dan sintesis senyawa ester dari biji durian beserta pengujian aktivitas antimikroba telah dilakukan terhadap bakteri patogen. Ekstraksi asam lemak dilakukan dengan metode maserasi menggunakan pelarut n-heksana. Sintesis metil ester asam lemak (MEAL) dan etil ester asam lemak (EEAL) dilakukan dengan menggunakan katalis H2SO4(p) dan NaOCH3. Senyawa hasil sintesis dianalisis menggunakan GC-MS dan FT-IR. Senyawa Asam Lemak (AL), MEAL, dan EEAL dengan variasi konsentrasi 10%, 20%, dan 30% diuji aktivitas antimikroba terhadap bakteri patogen S. Aureus, E. coli, S. Mutans dan C. Albicans dengan metode difusi cakram. Proses maserasi biji durian dengan n-heksana memberikan rendemen 3,45%. Komposisi minyak biji durian terdiri dari asam lemak bebas, trigliserida, digliserida, dan monogliserida. Kadar molekul trigliserida yang paling banyak adalah C50-POP 17,5878%; C54-SLS 14,5857% dan C50-PSP 12,7259%. Hasil sintesis dianalisis GC-MS, terdapat delapan senyawa ester yang diinterpretasi pola fragmentasinya yakni asam 10-oktadekadienoat metil ester, asam oktadekanoat metil ester, asam arakidonat metil ester, asam tetradekanoat etil ester, asam 9-heksadekadienoat etil ester, asam oktadekanoat etil ester, asam 9-oktadekadienoat etil ester dan asam arakidat etil ester. Uji aktivitas antimikroba menunjukkan bahwa aktivitas tertinggi terhadap C. Albicans ditunjukkan oleh MEAL 30% dengan indeks antimikroba (IAM) = 0,91. Sedangkan untuk S. Aureus, E. Coli dan S. Mutans aktivitas tertinggi ditunjukkan oleh AL 30% dengan IAM 0,75; 2,93 dan 1,38. Berdasarkan data diatas, senyawa AL dan esternya memiliki aktivitas sebagai antimikroba terhadap S. Aureus, E. Coli, S. Mutans, dan C. Albicans.
