dc.description.abstractBackground: Until now, the etiology and mechanism of SCF have not been fully known. But looking at it through histopathology, microvascular inflammation has been found. APN is an important modulator of inflammation that has multiple protection effects on the vascular endothelium. In SCF, there is low APN level that may contribute to the occurrence of SCF. Aim: To analyze on the polymorphism of APN genes and the APN level on Acehense patients with SCF. Method: All groups were examined on their APN levels using the ELISA, and PCR Polymorphism methods of ADIPOQ rs2241766, and rs1501299 were done using the TaqMan GTXpress Master Mix reagent (2x). Results: APN levels were lower in SCF and CAD compared to the control group, and were statistically significantly different. TIMI frames in the SCF group were higher than SCF, in which SCF was much involved in RCA while CAD were much involved in LAD. The analysis of APN polymorphism resulted in no difference in genotype and allele distribution either on +276G>T and +45T>G. There were significant differences in the levels of adiponectine genotype TT gene +45GT and GG genotype in the gene +276G>T in the three groups. Conclusion: This study shows that the APN levels are associated with the occurrence of CAD and SCF. The genotype analysis and APN gene alleles were found to be not correlated to the occurrence of CAD and SCF, but the TT genotype at +45T>G and GG at +276G>T was correlated to the APN levels.en_US
dc.description.abstractLatar Belakang : Sampai saat ini etiologi dan mekanisme ALK belum sepenuhnya diketetahui, namun secara histopatologi didapatkan adanya inflamasi mikrovaskuler. APN adalah modulator penting inflamasi yang mempunyai efek proteksi multiple pada endotel vaskuler dan pada ALK didapatkan kadar APN rendah yang mungkin berperan pada kejadian ALK. Tujuan : Untuk menganalisis Polimorfisme gen APN dan kadar APN pada pasien ALK pada suku Aceh. Metode : Semua kelompok dilakukan pemerikasaan kadar APN dengan metode ELISA dan PCR Polimorfime ADIPOQ rs2241766 dan rs1501299 menggunakan reagen TaqMan GTXpress Master Mix (2x) Hasil Penelitian : Kadar APN lebih rendah pada ALK dan PJK dibandingkan control dan secara statistik berbeda bermakna. TIMI frame pada kelompok ALK lebih tinggi dibandingkan PJK, pada ALK banyak terlibat RCA sementara PJK pada LAD. Analisis polimorfisme APN didapatkan tidak ada perbedaan distribusi genotype dan alel baik pada +276G>T dan +45T>G. Terdapat perbedaan bermakna kadar adiponektin genotype TT gen +45GT dan genotype GG pada gen +276G>T pada ketiga kelompok. Kesimpulan : Penelitian ini menunjukan, kadar APN berhubungan dengan kejadian PJK dan ALK. Analisis genotope dan alel gen APN didapatkan tidak berkorelasi dengan kejadian PJK dan ALK, namun genotype TT pada +45T>G dan GG pada +276G>T berhubungan dengan kadar APN.en_US
