    Pengaruh Ekstrak Umbi Bawang Dayak (Eleutherine bulbosa (Mill) Urb) Terhadap Kadar Glukosa Darah, Insulin dan Histopatologi Pankreas Tikus Wistar Diabetes yang Diberikan Diet Tinggi Lemak dan Diinduksi dengan Streptozotocin

    Abstract
    Latar belakang: Umbi bawang dayak (Eleutherine bulbosa (Mill) Urb) mengandung senyawa kimia yang diketehui berpotensi sebagai antidiabetik. Tujuan: Mengkaji pengaruh ekstrak etanol umbi bawang dayak (EEBD) terhadap perbaikan kondisi tikus diabetik yang diinduksi streptozotocin setelah pemberian diet tinggi lemak. Metode: Penelitian eksperimental dengan desain pre- dan post-test. Sebanyak 30 ekor tikus diabetik dibagi menjadi 5 kelompok dengan perlakuan satu kali sehari selama 14 hari: glibenklamid (Gli): 0,25 mg/kgbb, EEBD 125 mg/kgbb (BD125); 250 mg/kgbb (BD250); 500 mg/kgbb (BD500) dan CMC 1% bb (CMC). Evaluasi meliputi kadar glukosa darah (KGD) puasa, kadar insulin, gambaran histopatologi pulau langerhans. Data dianalisis dengan uji parametrik dan non parametrik. Hasil: KGD puasa menunjukkan penurunan yang signifikan setelah 7 jam pengamatan (p<0,001) pada tiap dosis, dan mulai hari ke-9 pada BD 500 (p<0,01) dengan penurunan KGD sebesar 187 mg/dL (50%). Setelah 14 hari perlakuan dijumpai penurunan KGD pada BD125 sebesar 206 mg/dL (61,6 %); BD250 sebesar 259 mg/dL (71,8%) dan BD500 sebesar 274 mg/dL (77,3%). Secara statistic penurunan ini berbeda secara signifikan dibandingkan dengan kelompok negatif (CMC) (p<0,001). Kadar insulin BD 250 meningkat bermakna (p<0,05). Gambaran histopatologi menunjukkan terjadinya kerusakan pada struktur pulau langerhans dan hanya BD500 yang menunjukkan adanya perbaikan. Dijumpai perbedaan luas yang bermakna pada BD250 dan BD 500 (p<0,05-0,01). Kesimpulan: EEBD mampu memperbaiki KGD, kadar insulin, struktur dan luas pulau Langerhans tikus diabetik
     
    Introduction: Dayak onion tuber extract (Eleutherine bulbosa (Mill) Urb) contained chemical compounds that has been known potentially as diabetic agent. Objective: This study aimed to determine dayak onion tuber extract (EEBD) on improvement of condition diabetic rats that were induced by streptozotocin after administration of high lipid diet. Method: The reaserch was designed experimentally using pre-test and post-test. 30 diabetic rats were divied into 5 group, once daily for 14 day as follows : glibenclamide (Gli): 0,25 mg/kgbb, EEBD 125 mg/kgbb (BD125); 250 mg/kgbb (BD250); 500 mg/kgbb (BD500) dan CMC 1% bb (CMC). Evaluations were included fasting blood glucose level (FBGL), insulin level, histopatology of islet of Langerhans. The data was analysed with parametric and non parametric statistic. Result: FBGL showed significant reduction at 7 hours observation (p<0,001) for each dose and at day 9 on BD500 (p<0,01) which decreased BGL 187 mg/dL (50%). After 14 dyas treatment, FBGL on BD125 was 206 mg/dL (61,6 %); BD250 was 259 mg/dL (71,8%), while BD500 was 274 mg/dL (77,3%). Statistically those reduction significantly different compared to negative control (CMC) (p<0,001, respectively). Insulin level of BD250 only was increased (P<0,05). The histopatology study showed only BD500 showed an improvement on the destruction of Langerhans’ structure. There were significant area large on BD250- and BD500-treated groups (p<0,05-0,01). Conclusion : EEBD able to improve blood glucose level, insulin level, structure and Langerhans area of diabetic rats.
