    Hubungan Antara Polimorfisme Gen INF-γ +874 A/T dan Skizofrenia pada Suku Batak

    Abstract
    Latar belakang Skizofrenia memiliki heritabilitas yang luas, identifikasi gen yang rentan terhadap risiko gangguan ini juga cukup sulit dibuktikan. Meskipun belum ada satu gen yang diidentifikasi khusus untuk skizofrenia, beberapa kandidat gen telah dikenal berhubungan dengan kerentanan terhadap skizofrenia serta memberi petunjuk untuk patofisiologi skizofrenia. Peran potensial kerentanan gen untuk pengembangan skizofrenia misalnya terlihat pada beberapa kompleks gen Tumor Necrosis Factor-alpha (TNF-alpha), Interleukin-1 (IL-1), Interleukin-6 (IL-6) dan Interleukin- 10 (IL-10).Beberapa peneliti menyebutkan keterlibatan Interferon Gamma (IFN-γ) dalam gangguan neuropsikiatri. Tujuan Mengetahui hubungan antara polimorfisme gen IFN-γ +874 A/T dan skizofrenia pada penderita skizofrenia suku Batak. Metode Studi ini adalah studi kasus kontrol yang dilakukan pada 248 subjek penelitian,124 subjek dalam kelompok kasus dan 124 subjek dalam kelompok kontrol akan diikutsertakan, Kelompok kasus adalah subjek berumur 20-60 tahun, dirawat inap di RSJ Prof. M. Ildrem Medan, Indonesia. Kelompok kontrol adalah pendonor di Unit transfusi darah Rumah Sakit Umum Pirngadi, Medan, Indonesia..Polimorfisme gen IFN-γ +874 A/T diperiksa dengan metode PCRRFLP. Hasil Frekuensi genotip AA pada kelompok pasien adalah 31,5%, pada kelompok kontrol adalah 46%. OR ditemukan 2,83 (95% CI 1,36-5,86, p=0,005). Frekuensi genotype AT dari gen IFN-γ +874 A/T lebih tinggi pada kelompok kasus jika dibandingkan dengan kelompok kontrol (43.5% vs 41.1%, p=0.097, OR=1,83 95% CI 0,89-3,79). Untuk alel, alel T terlihat lebih tinggi pada pasien dibandingkan dengan kelompok kontrol (46.0% vs 33.5%, p=0.006, OR=0.59 95% CI 0.41-0.85, p=0.006). Kesimpulan Ditemukan hubungan antara polimorfisme gen IFN-γ +874 A/T dan skizofrenia pada ODS pada suku Batak. Genotipe AA meningkatkan risiko skizofrenia sebesar 2.83 kali dibandingkan TT.
     
    Background Schizophrenia has broad heritability, genes that are susceptible to this problem are also quite difficult to prove. Several candidate genes have been known to be linked to schizophrenia and provide clues for the pathophysiology of schizophrenia. The potential gene include Necrosis Factor-alpha (TNF-alpha), Interleukin-1 (IL-1), Interleukin-6 (IL-6) and Interleukin-10 (IL-10). Some researchers report participating in Gamma Interferon (IFN-γ) in neuropsychiatric disorders Aim To find out the association between IFN-γ +874 A/T gene polymorphisms and schizophrenia in Batak ethnic population Methods A case control study was performed in 248 subjects ( 124 cases and 124 controls) aged between 20-55 years. Cases were those who were hospitalized in Professor Ildrem Mental Hospital, Medan, Indonesia. Controls were volenteer of blood donor in Pirngadi General Hospital of Medan, Indonesia. Polymorphism of IFN-γ +874 A/T gene was genotyped using PCR-RFLP method. Result The frequencies of genotype AA among patients was 31,5%, and 46% in control group (p=0.005, OR=2.83 95% CI1.36-5.86). Genotype AT of the IFN-γ +874 A/T was higher among patients in comparison with control group (43.5% vs 41.1%, p=0.097, OR=1.83 95% CI 0,89-3,79). In allelic frequencies, it is shown that T allele was higher in patient compare to control group (46.0% vs 33.5%, p=0.006, OR=0.59 95% CI 0.41-0.85, p=0.006). Conclusion We found an association between IFN-γ +874 A/T gene polymorphisms and schizophrenia in Batak population. The AA genotype was found to increase the risk of schizophrenia by 2.83 times, while the T allele was a protector of schizophrenia.
