dc.description.abstract | Backgound: Changes in lifestyle due to modernization and globalization tend to increase the risk of vascular diseases such as coronary heart disease and stroke. Currently one of the ischemic stroke therapies is tissue plasminogen activator (tPA). Various studies have been developed in stroke therapy that is more effective and still limited in neuroprotection to prevent the expansion of brain injuries. Various kinds of research on alternative medicine have been carried out. One that is related and an alternative medicine in the treatment of ischemic stroke is snakehead fish extract (Channa striata) or Ophiocephalus striatus.
Objective: To prove the effect of Channa striata extract on VEGF levels, NO levels and VEGFR2 expression in the cerebral angiogenesis process of Sprague-Dawley strain rat ischemic stroke model.
Methods: The study design was true experimental of 25 Sprague-Dawley strain male rats divided into 5 groups. In the control group K- without carotid artery ligation and without the administration of snakehead fish extract (Channa striata). Group of K+ with carotid artery ligation and without the administration of snakehead fish extract (Channa striata). Group of P1 with carotid artery ligation and snakehead fish extract (Channa striata) at a dose of 200 mg/day. Group of P2 with carotid artery ligation and snakehead fish extract (Channa striata) at a dose of 400 mg/day. Group of P3 with carotid artery ligation and snakehead fish extract (Channa striata) at a dose of 800 mg/day. The blood sample was taken on the third day to evaluate vascular endothelial growth factor (VEGF) and nitric oxide (NO) levels, as well as to evaluate vascular endothelial growth factor receptor 2 (VEGFR2) expression.
Results: There was a significant increase in VEGF levels in the administration of snakehead fish extract (Channa striata) compared to the control group K+ (p <0,05). There was a significant increase in NO levels in the administration of snakehead fish extract (Channa striata) compared with the control group K+ (p <0,05). There was a significant increase in VEGRF2 expression in the administration of snakehead fish extract (Channa striata) compared to the control group K+ (p <0,05).
Coclusions: The administration of snakehead fish extract (Channa striata) increases VEGF levels, NO levels and VEGRF2 expression in the cerebral angiogenesis process of Sprague-Dawley strain rat ischemic stroke model. | en_US |
dc.description.abstract | Latar Belakang: Perubahan pola hidup akibat modernisasi dan globalisasi pada dewasa ini cenderung meningkatkan risiko terjadinya penyakit vaskular seperti jantung koroner dan stroke. Saat ini salah satu terapi stroke iskemik yang diterima adalah tissue plasminogen activator (tPA). Berbagai penelitian telah banyak dikembangkan dalam terapi stroke yang lebih efektif dan masih terbatas pada bidang neuroproteksi untuk mencegah perluasan cedera otak. Berbagai macam penelitian mengenai alternative medicine telah banyak dilakukan. Salah satu yang berkaitan dengan hal tersebut dan menjadi alternative medicine dalam pengobatan stroke iskemik adalah ekstrak ikan gabus (Channa striata) atau Ophiocephalus striatus.
Tujuan: Membuktikan efek pemberian ekstrak Channa striata terhadap kadar VEGF, kadar NO dan ekspresi VEGFR2 dalam proses angiogenesis serebral tikus galur Sprague - Dawley model stroke iskemik.
Metodologi: Merupakan penelitian eksperimental murni pada 25 hewan coba tikus jantan galur Sprague - Dawley yang dibagi menjadi 5 kelompok. Pada kelompok kontrol K- tanpa ligasi arteri karotis dan tanpa pemberian ekstrak ikan gabus (Channa striata). Kelompok K+ dengan ligasi arteri karotis dan tanpa pemberian ekstrak ikan gabus (Channa striata). Kelompok P1 dengan ligasi arteri karotis dan ekstrak ikan gabus (Channa striata) dosis 200 mg/hari. Kelompok P2 dengan ligasi arteri karotis dan ekstrak ikan gabus (Channa striata) dosis 400 mg/hari. Kelompok P3 dengan ligasi arteri karotis dan ekstrak ikan gabus (Channa striata) dosis 800 mg/hari. Selanjutnya dilakukan pengambilan darah pada hari ketiga untuk pemeriksaan kadar vascular endothelial growth factor (VEGF) dan kadar nitric oxide (NO), serta pemeriksaan ekspresi vascular endothelial growth factor receptor 2 (VEGFR2).
Hasil: Terdapat peningkatan signifikan kadar VEGF pada kondisi pemberian ekstrak ikan gabus (Channa striata) dibandingkan dengan kelompok kontrol K+ (p<0,05). Terdapat peningkatan signifikan kadar NO pada kondisi pemberian ekstrak ikan gabus (Channa striata) dibandingkan dengan kelompok kontrol K+ (p<0,05). Terdapat peningkatan signifikan ekspresi VEGRF2 pada kondisi pemberian ekstrak ikan gabus (Channa striata) dibandingkan dengan kelompok kontrol K+ (p<0,05).
Simpulan: Pemberian ekstrak ikan gabus (Channa striata) dapat meningkatkan kadar VEGF, kadar NO dan ekspresi VEGRF2 pada proses angiogenesis serebral tikus galur Sprague - Dawley model stroke iskemik. | en_US |
