    Analisis Kemampuan Nacre dalam Ekspresi TGF- β1 Menginduksi Dentin Tertier ( In Vivo)

    Abstract
    The pulp capping material is applied to the reversible pulpitis can induce TGF-β1 to form tertiary dentin so that the pulp tissue remains in a vital state. The aim of the study was to evaluate the ability of natural material, the Nacre induced TGF-β1 to form tertiary dentin and to evaluated the difference in the ability of Biodentine with Nacre to induce tertiary dentin by observing TGF-β1 expression in the period of 7,14 and 30 days. The samples were 30 male Wistar rats (Rattus Novergicus) which were divided into 3 big groups. The three major groups were divided based on observations of 7 days, 14 days and 30 days after the application of the pulp capping materials. The maxillary molar teeth were prepared until the pulp roof perforated, then in the first treatment group was done direct pulp capping by Nacre and it was restorated with RMGIC, the second treatment group was done direct pulp capping by Biodentine and it was restorated with RMGIC, the control group was immediately restorated with RMGIC, then the rats were sacrificed after the 7 days, 14 days and 30 days for the teeth was taken then they were stained with Hematoxylin Eosin (HE) and Immunohistochemistry (IHC) , after that the observation was done by binocular microscopy to see the results of TGF-β1 staining on the pulp of the Wistar rat’s teeth by 400x magnification. The results of research were analyzed in a nonparametric way using the Kruskal-Wallis test, the Mann-Whitney test, the Friedman test and the Wilcoxon test. Based on this research that the conclusion was The Nacre was able to induce TGF-β1 to form tertiary dentin and there was no significant difference in the ability of Nacre with Biodentine in inducing tertiary dentin (p<0,05).
     
    Bahan kaping pulpa yang diaplikasikan pada pulpa yang mengalami pulpitis reversibel dapat menginduksi TGF-β1 membentuk dentin tertier sehingga jaringan pulpa tetap dalam keadaan vital. Penelitian ini bertujuan untuk mengetahui kemampuan bahan alam yaitu Nacre dalam menginduksi TGF-β1 membentuk dentin tertier dan untuk mengetahui perbedaan kemampuan Biodentine dengan Nacre menginduksi dentin tertier dengan mengamati ekspresi TGF-β1 pada periode 7,14 dan 30 hari. Sampel yang digunakan sebanyak 30 tikus wistar (Rattus Novergicus) jantan yang dibagi menjadi 3 kelompok besar. Tiga kelompok besar tersebut dibagi berdasarkan pengamatan waktu 7 hari, 14 hari dan 30 hari setelah aplikasi bahan kaping pulpa. Pada semua kelompok dilakukan preparasi gigi molar maksila sampai perforasi mencapai atap pulpa, kemudian pada kelompok perlakuan pertama dilakukan direct pulp capping dengan Nacre dan ditutup dengan RMGIC, kelompok perlakuan kedua dilakukan direct pulp capping dengan Biodentine dan ditutup dengan RMGIC, pada kelompok kontrol langsung ditutup dengan tumpatan RMGIC, kemudian tikus dikorbankan pada hari ke-7, 14 dan 30 untuk diambil giginya kemudian dilakukan pewarnaan dengan Hematoksilin Eosin (HE) dan pewarnaan Imunohistokimia (IHC), setelah itu dilakukan pengamatan dengan mikroskop binokuler untuk melihat hasil ekspresi TGF-β1 pada pulpa gigi tikus Wistar dengan pembesaran 400x. Data hasil penelitian dianalisis secara non parametrik dengan menggunakan uji Kruskal-Wallis , uji Mann-Whitney, uji Friedman dan uji Wilcoxon. Berdasarkan penelitian yang dilakukan dapat disimpulkan bahwa Nacre mampu menginduksi TGF-β1 untuk membentuk dentin tertier dan tidak ada perbedaan bermakna pada kemampuan Nacre dengan Biodentine dalam menginduksi dentin tertier (p<0,05).
