Introduction: The sikkam plant (Bischofia javanica Blume) is traditionally used as an anti-inflammatory and antibacterial. Sikkam is obtained compound of triterpenoids,tannins, glycosides, and flavonoids. Antiinflammatory drugs that are steroids and non-steroids have side effects such as stomach ulcers, osteoporosis, aggravating diabetes mellitus and muscle weakness. Because it needs to be done research to get alternative drugs. This study aimed to isolate and to determine the anti-inflammatory effects of active fraction of sikkam stem bark. Methods: Simplisia powder macerated used ethanol 80%, this ethanol extract was fractionated with n-hexane and ethylacetate. The extracts were examined for phytochemical screening and characterization. Antiinflammatory activity test was carried out on male rats using a plestymometer, which was divided into 14 groups, each group consist of five rats. Doses of ethanol extract, n-hexane fraction, etiasetate fraction and water fraction (residual) were 50, 100 and 200 mg/kgbw given to rats that were induced with λ carrageenan, as negative control was 0.5% CMC Na and positive control was diclofenac sodium. The most active fractions were separated using paper chromatography (KKt) with the silent phase of whatman paper and the mobile phase of BAA (n-butanol: acetic acid: water = 4: 1: 5), 1% HCl, and 15% acetic acid. Isolated were identified by ultraviolet (UV) and infrareed (IR) spectrophotometry. Results: The result of phytochemical screening of sikkam stem bark is obtained compound of triterpenoids/steroids, tannins, glycosides, and flavonoids. The characterization of the sikkam stem bark were water levels of 7,32; water soluble extracts 32,97; levels of soluble extract in ethanol 23,26; total ash content of 0,48; acid insoluble ash content 0,01. Ethanol extract, n-hexane fraction, and ethyl acetate fraction of sikkam stem bark gave anti-inflammatory activity for all doses. Ethanol extract and ethylacetate fraction at doses of 100 and 200 mg/kgbw from 240 to 360 minutes can significantly reduce the edema volume of rats feet with diclofenac sodium at 2.25 mg / kg (p ≥ 0.05). The highest percentage of inflammation inhibition was ethylacetate fraction of sikkam bark with a dose of 200 mg/kgbw. The isolation results obtained were Rf isolates; 0.71 (yellow) with a maximum wavelength of λ of 283.8 nm and obtained the -OH group, C = O, C = C, C = H and C-H aliphatic.
Conclusion: Ethanol extract, n-hexane fraction and ethyl acetate fraction had anti-inflammatory effect and the isolates obtained were flavonoid compounds. Latar belakang: Tumbuhan sikkam (Bischofia javanica Blume) secara tradisional telah digunakan sebagai antiradang dan antibakteri. Sikkam mengandung metabolit sekunder triterpenoid/steroida, tannin, glikosida, dan flavonoid. Obat antiinflamasi termasuk golongan steroid dan non-steroid yang memiliki efek samping seperti tukak lambung, osteoporosis, memperberat diabetes melitus, dan otot lemah. Karena itu dibutuhkan penelitian untuk alternative obat. Tujuan penelitian ini untuk mengisolasi dan uji aktivitas antiinflamasi fraksi aktif kulit batang sikkam (Bischofia javanica blume) terhadap tikus.
Metodelogi: Serbuk simplisia dimaserasi menggunakan etanol 80%, kemudian ekstrak etanol difraksinasi dengan pelarut n-heksan dan etilasetat, selanjutnya serbuk simplisia dan ekstrak etanol diskrining fitokimia dan karakterisasi. Uji aktivitas antiinflamasi dilakukan terhadap tikus jantan menggunakan alat plestymometer, yang dibagi menjadi 14 kelompok dengan masing-masing terdiri 5 ekor. Dosis ekstrak etanol , fraksi n-heksan, fraksi etiasetat dan fraksi air (sisa) adalah 50, 100 dan 200 mg/kg bb yang diberikan pada tikus yang telah diinduksi dengan λ karagenan, sebagai kontrol negatif Na CMC 0,5% dan kontrol positif natrium diklofenak. Fraksi paling aktif dipisahkan menggunakan kromatografi kertas (KKt) dengan fase diam kertas whatman dan fase gerak BAA (n-butanol: asam asetat: air = 4:1:5), HCl 1 %, dan asam asetat 15%,. Isolat diidentifikasi dengan spektrofotometri ultraviolet (UV) dan Infrareed (IR).
Hasil: Hasil skrining fitokimia serbuk simplisia dan ekstrak etanol kulit batang sikkam diperoleh senyawa triterpenoid/steroid, tanin, glikosida dan flavonoid. Hasil karakterisasi ekstrak etanol untuk kadar air 7,32; kadar sari larut air 32,97; kadar sari larut etanol 23,26; kadar abu total 0,48 dan kadar abu tidak larut asam 0,01. Hasil pengujian ekstrak etanol, fraksi n-heksan, fraksi etilasetat dan kulit batang sikkam memberikan aktivitas antiinflamsi untuk semua dosis. Ekstrak etanol dan fraksi etilasetat dengan dosis 100 dan 200 mg/kgbb dari menit ke 240 sampai 360 dapat menurunkan volume edema telapak kaki tikus yang signifikan dengan natrium diklofenak dosis 2,25 mg/kg (p ≥ 0.05). Persen inhibisi radang yang paling tinggi adalah fraksi etilasetat kulit batang sikkam dosis 200 mg/kgbb. Hasil isolasi diperoleh isolat Rf; 0,71 (berwarna kuning)dengan panajang gelombang λ maksimum 283,8 nm dan diperoleh adanya gugus -OH, C=O, C=C, C=H dan C-H alifatis.
Kesimpulan: Ekstrak etanol, fraksi n-heksan dan fraksi etilasetat dan fraksi air memiliki efek antiinflamasi dan isolat yang diperoleh adalah senyawa golongan flavonoid.
