    Sediaan Pelepasan Terkontrol: Kitosan sebagai Matrik Sediaan Tablet Press-Coating

    Abstract
    The chitosan had been isolated from the skin of swallow shrimp by deacetylation chitin, and then used as a matrix for the preparation of press-coating tablet. The press-coating tablet consists of core and coating, which chitosan was used as binder for the core and the coating of press-coating tablet. The excipient of core tablet contains only chitosan and lactose, but the coated tablet contains chitosan, lactose and Amylum Manihot. As a model drug was used furosemida, which was divided into the coated35% and 65% into the core. The results show that chitosan which is isolated from the skin of shrimps is suitable for characteristic of recipient and it can be applied a matrix for press-coating tablet of control release. The release of furosemide increases with increasing the concentration of chitosan in medium I (pH 1.2) and II (pH 5.8), but decrease with increasing of the certain concentration of chitosan on F-l, F-2 and F-3. The release of furosemide from press-coating tablet follows by the first order. The ideal concentration of chitosan as a matrix in the core tablet is about 15.38%. The release of furosemida in medium II (pH 5.8) and medium1lI (pH 7.4) are faster than in medium I (pH 1.2), but the release of furosemide in medium II (pH 5.8) is faster than in medium Ill (pH 7.4). The in vitro study of formula has correlation with the in vivo study with the value of R is about 0.8.
     
    Telah diteliti kitosan yang diisolasi dari kulit udang swallow dengan cara deasetilasi kitin, selanjutnya digunakan sebagai matrik sediaan tablet press-coating. Tablet press-coating terdiri dari dua bagian, yaitu bagian inti dan penyalut dimana kitosan digunakan sebagai bahan pengikat, baik pada inti maupun pada penyalut tablet. Bahan pembawa tablet inti hanya terdiri dari kitosan dan laktosa, sedangkan pada penyalut terdiri dati laktosa, kitosan dan amylum manihot. Sebagai model obat digunakan furosemida yangterbagipada penyalut35%dan pada inti 65%. Hasil penelitian menunjukkan bahwa kitosan hasil isolasi dari kulit udang swallow memenuhi karakteristik bahan tambahan dan dapat digunakan sebagai matrik sediaan pelepasan terkontrol tablet press-coating. Pelepasan furosemida semakin cepat dengan bertambahnya konsentrasi kitosan dalam medium I (pH 1,2), II (pH 5,8), tetapi pelepasan furosemida dalam medium III (pH 7,4) semakin lambat dengan bertambalmya konsetrasi kitosan tertentu untuk F-I, F-2, dan F-3. Kinetika pelepasan furosemida mengikuti orde satu, Konsentrasi kitosan yang ideal digunakan sebagai matrik dalam inti tablet press-coating adalah 15,38%. Pelepasan furosemida di da1am medium II (pH 5,8) dan medium III (pH 7,4) lebih cepat dibandingkan dengan medium I (pH 1,2), tetapi pelepasan furosemida dalammediumII (pH 5,8) lebih cepat dibandiagkan dengan pelepasan dalam medium III (pH 7,4). Formula yang diuji secara in vitro mempunyai korelasi dengan furmula yang diuji secara in Vivo dengan harga R mendekati 0,8.
