    Aktivitas Antidepresan Ekstrak Etanol Rimpang Temu Giring (Curcuma heyneana Val. & V): Peningkatan Aktivitas Lokomotor dan Penurunan Immobility Time pada Mencit

    Abstract
    Latar Belakang: Resistensi terapi terhadap depresi masih tinggi. Sehingga diperlukannya alternatif pengobatan lainnya. Curcuma heyneana termasuk ke dalam suku Zingiberaceae. Curcuma heyneana mengandung kurkumin yang diduga memiliki peran sebagai antidepresan. Tujuan: Untuk mengevaluasi aktivitas antidepresan Curcuma heyneana dapat menurunkan durasi immobility dan meningkatkan aktivitas lokomotor. Metode: Penelitian ini diawali dengan maserasi serbuk Curcuma heyneana, kemudian dilakukan karakterisasi dan skrining fitokimia serbuk dan ekstraknya. Chronic mild stress selama 14 hari digunakan untuk menginduksi depresi, diikuti dengan pemberian ekstrak dengan dosis 50, 100, 200 mg/kg BB selama 10 hari. Evaluasi antidepresan dilakukan dengan menggunakan tail suspension test (TST), forced swimming test (FST) untuk mengukur penurunan immobility time dan open field test (OFT) untuk mengukur peningkatan aktivitas lokomotor. Sertralin 6,5 mg/kg BB digunakan sebagai pembanding. Data dianalisis secara statistika menggunakan Analysis of Variance (ANOVA) dilanjutkan dengan uji post hoc. Hasil: Induksi depresi selama 14 hari dapat menyebabkan penurunan aktivitas lokomotor serta meningkatkan durasi immobility yang berbeda dengan kontrol normal (p<0,05). Hasil penelitian menunjukkan ekstrak dosis 100 dan 200 mg/kg BB memiliki rata-rata immobility time yang lebih rendah dari kontrol negatif (p<0,05) pada tail suspension test (<239,50 ± 9,67 detik) dan forced swimming test (<225,50 ± 16,95 detik) serta rata-rata yang lebih besar dari kontrol negatif (p<0,05) pada durasi central square (>6,25 ± 1,65 detik) dan jumlah crossing (>35,75 ±5,54 jumlah crossing). Kesimpulan: Ekstrak dosis 100, 200 mg/kg BB mempunyai aktivitas antidepresan dengan menurunkan durasi immobility, dan meningkatkan aktivitas lokomotor pada mencit.
     
    Background: The resistance of depression therapy still high. So that the other alternative therapy are needed. Curcuma heyneana included in Zingiberaceae family. Curcuma heyneana contains curcumin which is thought have an antidepressant. Objective: To evaluate the antidepressant activity of Curcuma heyneana by decrease the duration of immobility and increase the locomotor activity. Method: This research was started with maceration of Curcuma heyneana, then followed with characterization and phytochemical screening of powders and extracts. Chronic mild stres for 14 days is used to induce depression, followed by giving the extracts at dose 50, 100, 200 mg/kg BW for 10 days. Antidepressant evaluation was carried out using a tail suspension test (TST), forced swimming test (FST) to measure decrease immobility time and an open field test (OFT) to measure increase locomotor activity. Sertralin 6.5 mg / kg BW was used for comparison. The data were analyzed statistically using the Analysis of Variance (ANOVA) followed by the post hoc test. Result: Depression induction for 14 days can cause a decrease in locomotor activity and increase the duration of immobility, which is different from normal controls (p <0.05). The results showed that the extract at doses of 100 and 200 mg / kg BW had an average immobility time lower than the negative control (p <0.05) on the tail suspension test (<239.50 ± 9.67 seconds) and the forced swimming test. (<225.50 ± 16.95 seconds) and the average is greater than the negative control (p <0.05) on the duration of the central square (> 6.25 ± 1.65 seconds) and the number of crosses (> 35, 75 ± 5.54 number of crosses). Conclusion: Extract at the doses of 100 and 200 mg/kg BW have antidepressant activity by decreasing immobility time and increasing locomotor activity.
