    Kajian Sterilitas dan Analisis Residu Etilen Oksida di Dalam Alamat Kontrasepsi Dalam Rahim

    Abstract
    Sterility test and analysis of ethylene oxide residue in intra uterine device (IUD), a product of PT. Kimis Farma, Bandung had been carried out. Total microorganisms and thei group from unsterilized samples were determined using plate count and staining Gram methods Sterility test of sterilized samples were evaluated according to Farmukope Indonesia, edisi IV. Ethylene oxide residue in IUD was analyzed by gas chromatography with Porapak Q(R) column as stationary phase, nitrogen as mobile phase at flow rate of 30 mL/minute and 33 mL/minute and, flame ionization detector. Results' showed that detection, quantitation. and determination limit for ethylene oxide were 7.7; 22.38 and I ppm, respectively, while recovery and variance coefficient for ethylene oxide in IUD were 96% and 0.68% respectively. Total bacteria of unsterilized sample was 2.9 x 104 cfu/g, while Total fungi was 3.9 x 104 cfu/g, consisting of Bacillus and Sarcina and a group of fungi i.e. Aspergillus. All of the sterilized sample were sterile and ethylene oxide residue was not detected.
     
    Te1ah dilakukan kajian meliputi uji sterilitas dan analisis residu etilen oksida dalam alat kontrasepsi dalam rahim (AKDR) produksi PT. Kimia Farma, Bandung. Penentuan angka mikroba dan golongan mikroba dilakukan terhadap sampel yang tidak disterilkan dengan metode lempeng dan pewarnaan Gram. Uji sterilitas terhadap sampel yang sudah disterilkan dilakukan sesuai dengan, prosedur yang ada di Farmakope Indonesia, edisi IV. Analisis residu etilen oksida dilakukan dengan metode kromatografi gas, memakai kolom Porapak Q(R) sebagai fase diam, gas nitrogen murni sebagai fase gerak dengan kecepatan 30 mL/menit dan 33 mL/menit, dan detektor pengionan nyala. Hasil menunjukkan bahwa batas deteksi, batas kuantitasi, dan batas determinasi berturut-turut adalah 7,7; 22,38 dan 1 ppm, sedangkan perolehan kembali dan koefisien variansi untuk etilen oksida di dalam AKDR berturut-turut 96% dan 0,68%. AKDR yang belum disterilkan mempunyai angka total bakteri 2,9 x 10' cfu/g dan total kapang 3,9 x 104 cfulg, terdiri dari kelompok Bacillus dan Sarcina serta satu golongan kapang, yaitu Aspergillus. Semua sampel AKDR yang sudah disterilkan ternyata steril dan residu ctilen oksida di dalam AKDR tidak terdeteksi.
