    Korelasi Ekspresi Microrna-155 dengan Grade Histopatologi pada Jaringan Kanker Payudara Tipe Duktal

    Abstract
    MicroRNA (miRNA) is a short 18-22 nucleotide non-coding RNA that binds their target gene in the 3'-untranslated region (3'-UTR) causing direct mRNA degradation or repression of mRNA translation. MicroRNA has been known to be involved in myriads of biological processes including cellular proliferation, apoptosis, cell growth, cellular differentiation, and metabolism. Various miRNA has been identified to have changes in their expression in various types of cancers in human. This study aims to analyze the correlation between the expression of miR-155 and the histopathological grade of ductal type breast cancer. 64 parrafin tissue blocks from female breast cancer patients from the department of surgical oncology of Adam Malik Hospital between 21012-2014 were taken from the tissue bank of Anatomical Pathology Laboratory of Adam Malik Hospital and included in this crosssectional study. RNA from the blocks were isolated and the RNA quantity and quality were determined with spectrophotometer and followed by cDNA synthesis. Expression analysis of miR 155 was done using Real Time qPCR. The histopathological data were obtained from patient’s medical records. Spearman correlation test were used to analyze the correlation between expression of miR-155 and the histopathological grade. No significant correlation was found between the expression of miR-155 and the histopathological grade of the ductal type breast cancer tissues (p=0,300; r= -0,67), but the miR-155 expression from all the breast cancer tissues was higher than the UniSp6 RNA spike-in (internal control), and the highest expression was in the grade I breast cancer patients. Changes in miR-155 expression can be a potential biomarker for early diagnosis and prognostic predictor of breast cancer
     
    MicroRNA (miRNA) merupakan noncoding RNA pendek yang terdiri dari sekitar 18-22 nukleotida yang mengikat target gen nya di 3'-untranslated region (3'-UTR), menyebabkan degradasi langsung mRNA atau represi translasi mRNA. MicroRNA telah terbukti terlibat dalam pengaturan spektrum yang luas dari proses biologis, termasuk proliferasi sel, apoptosis, perkembangan sel, diferensiasi sel dan metabolisme. Berbagai miRNA telah diidentifikasi mengalami perubahan ekspresi dalam berbagai jenis kanker pada manusia. Penelitian ini bertujuan untuk menganalisis korelasi antara ekspresi miR-155 dengan grade histopatologi pada jaringan kanker payudara tipe duktal. 64 blok parafin jaringan kanker payudara dari perempuan pasien bedah onkologi di RSUP H. Adam Malik Medan tahun 2012-2014 yang diambil dari bank blok Laboratorium Patologi Anatomi RSUP H. Adam Malik Medan diikutsertakan pada penelitian cross sectional ini. RNA sampel diisolasi kemudian dicek kualitas dan kuantitas RNA hasil isolasi menggunakan spektrofotometer, dilanjutkan dengan sintesis cDNA dari RNA hasil isolasi, dan kemudian dilakukan analisis ekspresi miR-155 dengan menggunakan Real Time qPCR. Data grade histopatologi didapat dari data rekam medis pasien. Uji korelasi Spearman digunakan untuk menganalisis apakah dijumpai korelasi yang bermakna antara ekspresi miR-155 dengan grade histopatologi kanker payudara. Tidak dijumpai adanya korelasi yang bermakna antara ekspresi miR-155 dengan grade histopatologi pada jaringan kanker payudara tipe duktal (p=0,300; r=-0,67), tetapi ekspresi miR-155 pada seluruh jaringan kanker payudara lebih tinggi dibandingkan dengan UniSp6 RNA Spike-in (kontrol internal), dengan peningkatan ekspresi miR-155 yang tertinggi pada pasien kanker payudara grade I. Perubahan ekspresi miR-155 dapat menjadi calon biomarker yang potensial untuk diagnosis dini dan juga penentuan prognosis kanker payudara.
