Formulasi Bentuk Sediaan Sachet Nata De Coco yang Mengandung Piroksikam di Uji secara Invitro dan Uji Pencegah Ulser

Abstract

The research of formulation dosage form of nata de coco sachet containing a piroxicam testing by in vitro and testing ulser prevention was carried out. Dosage form of nata de coco sachet was made from nata de coco with size 11 x 11 cm, which containing piroxicam 20 mg. The dosage form of nata de coco sachet was made in three forms sachet, i.e : 1. without pore, 2. one pore, 3. two pores. The result of dissolution test in medium pH 1.2 and in the medium pH 7.4 by using ANOVA test showed that the rate of dissolution was significantly different (P<0.05) from powder sachet nata de coco without pore, one pore and two pores. Since the ANOVA test result was significantly different so that the test followed by LSD test. The LSD test result showed that the rate of dissolution in the medium of pH 1.2 there was significantly different between the dosage form of powder piroxicam and nata de coco sachet without pore and one pore, where as nata de coco sachet without pore and one pore was not significantly different. However the rate of dissolution in the medium pH 7.4 was significantly different from all the dosage form had tested. Testing of prevention ulser test using rabbit as experimental animal was carried out. The result of observation from rabbit’s gastric showed that the dosage form of nata de coco sachet without pore or nata de coco sachet one pore and two pores were no effect on gastric irritation rabbit.

Telah dilakukan penelitian tentang formulasi bentuk sediaan sachet nata de coco yang mengandung piroksikam di uji secara invitro dan uji pencegah ulser. Bentuk sediaan sachet nata de coco dibuat dari membran nata de coco berukuran 11x 11cm, yang mengandung 20 mg piroksikam. Sediaan sachet nata de coco tersebut dibuat dalam tiga bentuk sachet yaitu : 1. tanpa pori, 2. berpori satu,dan 3. berpori dua. Hasil uji disolusi dalam medium pH 1,2 dan medium pH 7,4 menggunakan uji ANOVA menunjukkan bahwa terdapat perbedaan kecepatan pelepasan yang signifikan (P< 0,05) baik pada medium pH 1,2 maupun medium pH 7,4, dari sediaan serbuk sachet nata de coco tanpa pori, berpori satu maupun berpori dua. Berhubung dari hasil uji ANOVA terdapat perbedaan yang signifikan, maka uji dilanjutkan dengan uji LSD. Hasilnya menunjukkan bahwa pada medium pH 1,2, terdapat perbedaan kecepatan pelepasan yang signifikan antara bentuk sediaan serbuk piroksikam dengan sachet nata de coco tanpa pori dan nata de coco berpori satu. Sedangkan pada sachet nata de coco tanpa pori dan berpori satu tidak berbeda secara signifikan. Pada uji LSD pada medium pH 7,4 terdapat perbedaan yang signifikan dari semua sediaan yang diuji ((P<0,05) Pengujian terhadap uji pencegah ulser dilakukan dengan menggunakan kelinci sebagai hewan percobaan . Dari hasil pengamatan pada lambung kelinci didapatkan bahwa sediaan sachet nata de coco tanpa pori atau dengan sachet nata de coco berpori satu,dan sachet nata de coco berpori dua ternyata tidak memberikan efek iritasi terhadap lambung kelinci.
