Pengaruh Ekstrak Buah Morinda Citrifolia Linn terhadap Kualitas , Kuantitas Sperma dan Kadar Malondialdehyde Testis Tikus Wistar Diabetes Mellitus

Abstract

Hyperglycemia in Diabetes Mellitus causes increased levels of free radicals. Autooxydation in the process of hyperglycemia triggers the formation of free radicals. Free radicals can damage cell membranes, a lipid peroxide or Malondialdehyde (MDA), if was continued lead to system damage cell membranes and cell death. Morinda Citrifolia Linn contain secondary metabolic compounds that contain nutrients that are also diverse as Vitamins A, C, Niacin, Thiamine and Riboflavin, and minerals such of iron, calcium, sodium, and potassium. Several types of phytochemical compounds in the noni fruit is alkaloids among others Xeronin and Proxeronine (precursors xeronine). Xeronine is an alkaloid which the human body needs to active the enzyme as well as organize and create to a protein structures. The purpose of this study was to the determine the effect of extract of the Morinda Citrifolia Linn fruits of Quality, Quantity of Sperm and Testicular Levels of Malondialdehyde of the Wistar Rats Diabetes Mellitus. Research subject was adult male Wistar rats with age ± 3 months, 150 – 200 grams, as much as 28 rats were divided into 4 groups: (1) P0 as control, (2) P1 = group with aloxan induction for 10 days, (3) P2 = group with 10 days of induction aloxan + extract of Morinda Citrifolia Linn for 18 days, and (4) P3 = group with the extract of Morinda Citrifolia Linn for 18 days + induction aloxan for 10 days. Count of sperm calculate by the Improved Neubauer Chamber Calculate and light microscopes. Test of MDA levels with a spectrophotometer. Ethical Clearance was obtained from the Animal Research Ethics Committee FMIPA USU. From the results obtained by increasing the quality and quantity of sperm in adult male rats with Diabetes Mellitus who received the extract of Morinda Citrifolia Linn compared with the group that no receive the extract Morinda Citrifolia Linn, but not significantly (p>0,05). Normal sperm morphology and sperm motility of adult male rats Diabetes Mellitus increases in group P3, but not significantly (p>0,05). Decline in testicular MDA levels are not significant (p>0,05). The increase in blood sugar levels in P3 group was significantly different (p<0,05) at which a given group P3 Morinda Citrifolia Linn can not decrease of blood sugar levels of adult male rats with Diabetes Mellitus

Hiperglikemia pada DM menyebabkan kenaikan kadar radikal bebas. Adanya proses autooksidasi pada hiperglikemi memicu pembentukan radikal bebas. Radikal bebas dapat merusak membran sel, menjadi lipid peroksida atau Malondialdehyde (MDA), bila berlanjut mengakibatkan kerusakan sistem membran sel dan kematian sel. Morinda Citrifolia Linn mengandung zat mengandung senyawa metabolik sekunder yang mengandung nutrisinya yang juga beragam seperti vitamin A, C, Niasin, Tiamin dan Riboflavin, serta mineral seperti zat besi, kalsium, natrium, dan kalium. Beberapa jenis senyawa fitokimia dalam buah mengkudu adalah alkaloidnya antara lain Xeronin dan Proxeronin (prekursor xeronin). Xeronin merupakan alkaloid yang dibutuhkan tubuh manusia untuk mengaktifkan enzim serta mengatur dan membentuk struktur protein. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak buah Morinda Citrifolia Linn terhadap Kualitas , Kuantitas Sperma dan Kadar Malondialdehyde Testis Tikus Wistar Diabetes Mellitus. Subjek penelitian Tikus Wistar jantan dewasa dengan usia ± 3 bulan, 150 – 200 gram, sebanyak 28 ekor dibagi dalam 4 kelompok yaitu (1) P0 sebagai kontrol, (2) P1 = kelompok dengan induksi aloksan selama 10 hari, (3) P2 = kelompok dengan induksi aloksan 10 hari + pemberian ekstrak Morinda Citrifolia Linn 18 hari, dan (4) P3 = kelompok dengan pemberian ekstrak Morinda Citrifolia Linn 18 hari + induksi Aloksan 10 hari. Pemeriksaan sperma dengan Kamar Hitung Improved Neubauer dan Mikroskop cahaya. Pemeriksaan kadar MDA dengan spektrofotometer. Ethical Clearance di peroleh dari Komisi Etik Penelitian Hewan FMIPA USU. Dari hasil diperoleh peningkatan kualitas dan kuantitas sperma pada tikus jantan dewasa diabetes mellitus yang mendapat ekstrak Morinda Citrifolia Linn dibandingkan dengan kelompok yang tidak mendapat ekstrak Morinda Citrifolia Linn, namun tidak nyata (p>0.05). Morfologi sperma normal dan motilitas sperma tikus jantan dewasa Diabetes Mellitus didapatkan peningkatan pada kelompok P3, namun tidak nyata (p>0.05). Terjadi penurunan kadar MDA testis yang tidak nyata (p>0.05). Kenaikan kadar gula darah pada kelompok P3 berbeda nyata (p<0.05) dimana pada kelompok P3 yang diberikan Morinda Citrifolia Linn tidak dapat mengurangi kadar gula darah tikus jantan dewasa Diabetes Mellitus
