dc.description.abstract | A study of chitinolytic bacteria from hot springs and characterization of its chitinase has been done. Isolation was done using minimum salt agar with colloidal chitin as the sole carbon source incubated at 53 °C for 7 days. Examination of chitinase was performed on the same medium. Precipitation of crude chitinase was performed with different levels of ammonium sulphate. Characterization of chitinase including pH, temperature and Km dan Vmax was carried out. Ten chitinolytic isolates were isolated from hot springs. JP2 showed more to utilize chitin with chitinase index of 1.65. The maximum activity of chitinase was produced 4 days after incubation with chitinase activity of 0.0063 U/ml. The maximum activity was 0.0087 U/ml at 50% of ammonium sulphate with specific activity of 0.046 U/mg. Chitinase activity increased at pH 6 and at 60 ᴼC by 0.05066 U/ml with specific activity 3.8113 U/mg after dialysis. Km and Vmax values were 0.321 mg/ml and 71.429 μg, respectively. Based on 16S rRNA gene identification, JP2 showed that isolate was Bacillus licheniformis. | en_US |
dc.description.abstract | Penelitian tentang penapisan bakteri kitinolitik dari sumber air panas Penen dan karakterisasi kitinasenya telah dilakukan. Isolasi dilakukan menggunakan medium agar garam minimum dengan kolidal kitin sebagai satu-satunya sumber karbon yang diinkubasi pada suhu 53 C selama 7 hari. Uji indeks kitinase dilakukan pada media yang sama, dengan melihat zona bening terbesar yang dihasilkan oleh masing-masing isolat. Presipitasi enzim kasar dilakukan dengan kadar amonium sulfat yang berbeda. Karakter kitinase yang dipelajari mencakup pH, suhu, dan nilai Km dan Vmaks. Sepuluh isolat kitinolitik diperoleh dari hasil isolasi. Isolat JP2 memiliki indeks kitinolitik tertinggi yakni 1,65. Produksi kitinase tertinggi isolat JP2 diperoleh pada hari ke-4 dengan aktivitas kitinase 0,0063 U/ml. Aktivitas tertinggi diperoleh pada konsentrasi ammonium sulfat 50% yakni sebesar 0,0087 U/ml dengan aktivitas spesifik sebesar 0,046 U/mg. Aktivitas kitinase meningkat secara signifikan pada pH optimum 6 dan suhu optimum 60 °C yakni 0,05066 U/ml (3,8113 U/mg) setelah dialisis. Nilai Km dan Vmaks adalah masing-masing 0,321 μg/ml dan 71,429 μg. Berdasarkan identifikasi gen 16S rRNA, menunjukkan bahwa isolat JP2 tergolong ke dalam Bacillus licheniformis. | en_US |
