dc.description.abstract | Background: the use antidepressants is often considered expensive, not easily available, and has fatal side effects. Several studies have reported that curcumin, one of the compounds contained in temu giring (Curcuma heyneana) can exert antidepressant-like effects in animal models commonly used to test the effect of antidepressants. Coffee (Coffea canephora) is also thought to have the potential to have antidepressant activity, such as increased locomotor activity.
Objective: to determine the increase in the antidepressant effect of the combination of ethanol extract of Coffea canephora (EECC) seed and Curcuma heyneana (EECH) rhizome compared to the single extract.
Method: this study used 6 treatment groups, namely sodium-Carboxymethyle Cellulose (CMC-Na) 0.5% as negative control, ethanol extract of Coffea canephora seeds (EECC) and ethanol extract of Curcuma heyneana rhizome (EECH) (1:1) in a dose of 50, 100, 200 mg / kgBW, sertralin 6.5 mg / kgBW as positive control, and normal control with ad libitum drinks, the duration of administration was 10 days. The method used to trigger depression is the chronic mild stress (CMS) method for 14 days. The antidepressant test method used is the tail suspension test (TST) and forced swimming test (FST) to measure the duration of immobility time, while open field test (OFT) was used to measure the duration of central squre (CS), the number of crosses, and grooming. Data were analyzed statistically using the Statistical Product and Service Solution (SPSS) program, followed by analysis of variance (ANOVA) test then the post hoc LSD test with a 95% confidence level.
Result: the combination of EECC and EECH (1:1) in a dose of 100 and 200 mg/kgBW significantly (p <0.005) reduced immobility time (IT) with a mean of 93.00 ± 19.61 and 84.25 ± 39.28 seconds on the TST method , then 159.50 ± 23.71 and 119.25 ± 18.6 seconds on the FST method. While the doses of 50, 100, 200 mg / kgBW significantly (p <0.005) increased locomotor activity on OFT with a mean CS of 27.00 ± 2.58, 31.00 ± 6.87, and 34.00 ± 4.71 seconds. The mean number of crosses was 80.25 ± 14.35, 98.75 ± 19.44, and 131.75 ± 14.53 times. The mean duration of grooming was 22.33 ± 10.48, 12.67 ± 4.06, and 9.67 ± 1.86 seconds. But the results of decreased IT duration and increased locomotor activity did not differ significantly when compared to the single extract. Conclusion: the combination of robusta coffee bean ethanol extract (Coffea canephora) and temu giring rhizome (Curcuma heyneana) (1: 1) in a dose of 50mg / kgBB, 100mg / kgBB, 200mg / kgBW cannot provide a significant increase in antidepressant effects compared to the single extract. | en_US |
dc.description.abstract | Latar Belakang: penggunaan antidepresan seringkali dianggap mahal, tidak mudah didapat, dan memiliki efek samping yang fatal. Beberapa penelitian telah melaporkan bahwa kurkumin, salah satu senyawa yang terkandung didalam temu giring (Curcuma heyneana) dapat memberikan efek seperti antidepresan pada model hewan yang biasa digunakan untuk melakukan pengujian efek antidepresan. Kopi (Coffea canephora) juga diduga berpotensi mempunyai aktivitas antidepresan seperti peningkatan aktivitas lokomotor.
Tujuan: untuk mengetahui peningkatan efek antidepresan kombinasi ekstrak etanol biji Coffea canephora (EECC) dan rimpang Curcuma heyneana (EECH) dibandingkan ekstrak tunggalnya.
Metode: penelitian ini menggunakan 6 kelompok perlakuan yaitu natrium-Carboxymethyle Cellulose (CMC-Na) 0,5% sebagai kontrol negatif, ekstrak etanol biji Coffea canephora (EECC) dan ekstrak etanol rimpang Curcuma heyneana (EECH) (1:1) dalam dosis 50, 100, 200 mg/kgBB, sertralin 6,5 mg/kgBB sebagai kontrol positif, kontrol normal dengan makanan minuman ad libitum, dan durasi pemberiaan selama 10 hari. Metode yang digunakan untuk memicu depresi yaitu metode chronic mild stress (CMS) selama 14 hari. Metode uji antidepresan digunakan tail suspension test (TST) dan forced swimming test (FST) untuk mengukur durasi immobility time, sedangkan open field test (OFT) untuk mengukur durasi central square (CS), jumlah cross, dan grooming. Data dianalisis secara statistik menggunakan program Statistical Product and Service Solution (SPSS), dilanjutkan uji analysis of variance (ANOVA) kemudian uji Post Hoc LSD dengan tingkat kepercayaan 95%.
Hasil: kombinasi EECC dan EECH (1:1) dalam dosis 100 dan 200 mg/kgBB signifikan (p<0,005) mengurangi immobility time (IT) dengan rerata 93,00 ± 19,61 dan 84,25 ± 39,28 detik pada metode TST, lalu 159,50 ± 23,71 dan 119,25 ± 18,6 detik pada metode FST. Sedangkan dosis 50, 100, 200 mg/kgBB signifikan (p<0,005) meningkatkan aktivitas lokomotor pada OFT dengan rerata CS 27,00 ± 2,58, 31,00 ± 6,87, dan 34,00 ± 4,71 detik. Rerata jumlah crossing 80,25 ± 14,35, 98,75 ± 19,44, dan 131,75 ± 14,53 kali. Rerata durasi grooming 22,33 ± 10,48, 12,67 ± 4,06, dan 9,67± 1,86 detik. Tetapi hasil penurunan durasi IT dan peningkatan aktivitas lokomotor tidak berbeda secara signifikan bila dibandingkan dengan ekstrak tunggalnya.
Kesimpulan: kombinasi ekstrak etanol biji kopi robusta (Coffea canephora) dan rimpang temu giring (Curcuma heyneana) (1:1) dalam dosis 50mg/kgBB, 100mg/kgBB, 200mg/kgBB tidak dapat memberikan peningkatan efek antidepresan yang signifikan dibandingkan dengan ekstrak tunggalnya. | en_US |
