    Abstract
Pulpal injury will cause a local inflammatory response that activates neural systems of the pulp (A delta and C) to release the neural proteins (neuropeptides) among which substance P (SP) is a neuropeptide that plays important role at the beginning of inflammation. SP is the first line of inflammatory mediator that are vasodilators and an algogenic substance and a strong chemotactic agent so that the its increase in concentration in the pulp tissue can cause changes in blood flow rate and elevation of pulpal blood pressure. Initial process of tissue mineralization is characterized by increased concentrations of alkaline phosphatase (ALP) as a marker (marker) reparative dentinogenesis. Watermelon Frost is natural remedy derived from watermelon (Citrullus vulgaris) used by Chinese community in reducing inflammation and promoting healing process. This study was aimed to examine the effect of watermelon frost on the expression of SP and ALP in reversible pulpitis. The subjects in this study were 32 teeth of Macacafascicularis that were given mechanical injury until the pulp was exposed. Group I: Induction of reversible pulpitis is done mechanically by drilling method. Pulp tissues were excavated and stored at -80 ° C until tested by ELISA; Group II: Reversible pulpitis applied with watermelon frost and teeth extraction was performed after 3 (three) days and pulp tissues were obtained by breaking the tooth. Pulp tissues were stored at -80 ° C until tested by ELISA. Bootstrapping and t test showed the mean and SD values in group I are 159.04 ± 7:35 (SP) and 38 133 ± 3.2 (ALP), while the second group are 45 361 ± 3.2 (SP) and 143.26 ± 9:36 (ALP). Paired t test in each group showed significant differences in SP and ALP concentrations between two groups (p <0.0001). The results of this study showed that watermelon frost has effect on the decrease in SP expression and increased expression of ALP in reversible pulpitis.
