    Pengaruh Pemberian Ekstrak Etanol Kulit Buah Manggis (Garcinia Mangostana.L) terhadap Perubahan Makroskopis, Mikroskopis dan Tampilan Immunohistokimia Antioksidan Copper Zinc Superoxide Dismutase (Cu Zn SOD) pada Ginjal Mencit Jantan (Mus Musculus.L) Strain DDW yang di Papari Oleh Monosodium Glutamate (MSG) Dibandingkan Dengan Vitamin E

    Abstract
    Introduction : Extract of Mangosteen rind (Garcinia Mangostana L) is widely used as a medicine, one of them as antioxidants to prevent the effect of free radicals. The use of MSG as a flavoring in food have been reported have a toxic effects in the kidney caused decreased kidney function and increased lipid peroxidation leading to oxidative stress. Giving ethanolic extract of mangosteen rind orally sugested reduce effects of free radicals. This study aimed to know the effectiveness ethanolic extract of mangosteen rind as antioxidants in mice given MSG. Method : This is an experimental investigation, using mice. The mice divided into 5 groups. First group (P0) was a control group only given distilled water 0.3 ml/mouse/day on 1-35 days, (P1):was given MSG 8 mg/g bw/day on 1-21 days and 22-35 days no treatment. (P2): was given MSG 8 mg/g bw/day on 1-21 days and 22-35 days given ethanolic extract of mangosteen rind 600 mg/kg bw/day, (P3): was given MSG 8 mg/g bw/day on 1-21 days and on 22-35 days of vitamin E 0.2 mg/g bw/day, and (P4):only given mangosteen rind extracts as much as 600 mg/kg bw/day for 14 days. Result : The results showed MSG in the given dose produced macroscopic damage to the kidney although not significant (p>0.05), but microscopically significant (p<0.05) and with ethanolic extract of mangosteen rind could be repaired significantly (p<0,05). MSG could suppress the immunohistochemistry appearance of copper zinc superoxide dismutase (Cu Zn SOD) in the kidney significantly (P<0.05), but with ethanolic extract of mangosteen rind this appearance improved significantly (p<0,05). Conclusion: In this experiments, it was found that the ethanolic extracts of the mangosteen rind could repair the damage in the kidney induced by MSG, microscopicallay and also improved the immunohistochemistry appearance .
     
    Pendahuluan : Ekstrak kulit manggis (Garcinia Mangostana L) yang selama ini telah banyak digunakan masyarakat sebagai obat, salah satunya sebagai antioksidan untuk mencegah radikal bebas. Penggunaan MSG sebagai penyedap dalam makanan telah dilaporkan mempunyai efek toksik terhadap ginjal yang dapat menurunkan fungsi ginjal dan menyebabkan peningkatan peroksidasi lipid sehingga menimbulkan stres oksidatif. Pemberian EEKM secara oral diharapkan dapat menurunkan efek radikal bebas. Penelitian ini bertujuan untuk mengetahui efektifitas EEKM sebagai antioksidan pada mencit yang dipapari dengan MSG. Metode : Metode penelitian yang digunakan adalah metoda eksperimental yang dilakukan terhadap mencit jantan dengan 5 kelompok perlakuan dan 5 ulangan. Kelompok (P0): kontrol aquadest 0,3 ml/mencit/hari dari hari 1-35, (P1): MSG 8 mg/g BB mencit/hari dari hari 1-21 dan hari ke 22-35 tanpa perlakuan. (P2): MSG 8 mg/g BB mencit/hari dari hari 1-22 dan hari ke 22-35 pemberian EEKM 600mg/kg BB mencit/hari, (P3): MSG 8 mg/g BB mencit/hari dari hari 1-21 dan pada hari ke-22-35 pemberian vitamin E 0,2 mg/g BB mencit/hari, dan (P4): EEKM sebanyak 600 mg/kg BB mencit/hari selama 14 hari. Hasil penelitian : Hasil penelitian menunjukkan pemberian MSG menyebabkan kerusakan makroskopis ginjal walaupun secara tidak bermakna (p>0,05), namun menyebabkan kerusakan mikroskopis pada ginjal secara bermakna (p<0,05) dan dapat diperbaiki dengan pemberian EEKM secara bermakna (p<0,05). MSG dapat menurunkan tampilan immunohistokimia antioksidan copper zinc superoxide dismutase (Cu Zn SOD) pada ginjal secara bermakna (p<0,05), namun tampilan immunohistokimia copper zinc superoxide dismutase (Cu Zn SOD) ini dapat ditingkatkan secara bermakna dengan pemberian EEKM (p<0,05).
