| dc.description.abstract | Colon cancer is the third leading cause of death from cancer. One of strategy colon cancer treatment based on the discovery of cytotoxic agents is now being developed. Mangrove is the plants producing secondary metabolite compounds mainly from the group of isoprenoid compounds. Polyisoprenoids (dolichol and polyprenol) is known to have benefits as antimicrobial, anticancer, and antiviral. Therefore, this study aims to determine the effects of n-hexane fraction from seven mangrove: Putut (Bruguiera gymnorrhizaI), Tengar (Ceriops tagal), Nipah (Nypa fruticans), Bakau Hitam (Rhizophora mucronata), Pandan Laut (Pandanus odoratatissima), Malapari (Pongamia pinnata),and Jarak (Ricinus communis) based on the distribution and diversity of polyisoprenoids compounds against cytotoxic activity of colon cancer cells.
Effects on cell proliferation and cell death were assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay method. The mechanism of actions of the active fraction were measured by apoptosis and cell cycle were assesed by flow cytometric, and the expression of Bcl-2 and Cyclin D1 were observed by mmunocytochemistry method.
Results showed that n-hexane fraction of nipah, tengar, and bakau hitam leaf had the highest anticancer activity with an IC50 of 180,086 μg/mL, 276,055 μg/mL, and 278,335 μg/mL. Polyisoprenoid of nipah, tengar, and bakau hitam leaves decreased significantly the gene expression of Bcl-2 and Cyclin D1, and induced apoptosis and cell cycle arrest.
This study has evaluated the anticancer activity of n-hexane extract from nipah, tengar, and bakau hitam leaves have the potential to be used as a source of anticancer agents. N-hexane from nipah, tengar, and bakau hitam leaves against colon cancer cells and provided a basis of further development of the herbal extract for treatment of colon cancer. | en_US |
| dc.description.abstract | Kanker kolon merupakan penyebab utama kematian ketiga akibat kanker. Salah satu strategi pengobatan kanker kolon berdasarkan penemuan agen sitotoksik kini sedang dikembangkan. Mangrove merupakan tanaman penghasil senyawa metabolit sekunder terutama dari golongan senyawa isoprenoid. Polyisoprenoids (dolichol dan polyprenol) diketahui memiliki manfaat sebagai antimikroba, antikanker, dan antivirus. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak n-heksan dari tujuh spesies mangrove yaitu: Putut (Bruguiera gymnorrhiza), Tengar (Cariops tagal), Nipah (Nypa fruticans), Bakau Hitam (Rhizophora mucronata), Pandan Laut (Pandanus odoratatissimus), Malapari (Pongamia pinnata), dan Jarak (Ricinus communis) berdasarkan distribusi dan keragaman senyawa polyisoprenoids terhadap aktivitas sitotoksik sel kanker kolon (WiDr).
Efek proliferasi sel dan kematian sel dilakukan dengan metode 3- (4,5-dimetilthiazol-2-yl) -2,5-diphenyltetrazolium bromide. Ekstrak yang paling aktif dalam menyebabkan kematian sel WiDr dilanjutkan untuk pengujian terhadap apoptosis dan siklus sel dengan metode flow sitometri. Ekspresi Bcl-2 dan Cyclin D1 diamati dengan metode imunositokimia.
Hasil penelitian menunjukkan bahwa ekstrak n-heksan daun Nipah (Nypa fruticans), tengar (Ceriops tagal), dan bakau hitam (Rhizophora mucronata) memiliki aktivitas antikanker tertinggi dengan nilai IC50 berturut-turut 180,086 μg/mL, 276,055 μg/mL, dan 278,335 μg/mL. Ekstrak n-heksan dari daun nipah, tengar, dan bakau hitam mampu memacu apoptosis, dan menghambat siklus sel pada fase G0/G1, dengan mekanisme menekan ekspresi gen Bcl-2 dan Cyclin D1.
Aktivitas antikanker ekstrak n-heksan dari daun nipah, tengar, dan bakau hitam berpotensi untuk digunakan sebagai sumber agen antikanker. Ekstrak n-heksan dari daun nipah, tengar, dan bakau hitam bersifat sitotoksik pada sel kanker kolon (WiDr) dan memberikan dasar pengembangan lebih lanjut untuk pengobatan kanker kolon. | en_US |
