    Perbandingan Kadar Enzim Glutatione Peroxidase Lensa pada Penderita Katarak Senilis Perokok dan Bukan Perokok

    Abstract
    Introduction: The lens of the eye is very sensitive to the occurrence of oxidative stress, so many theories that developed about the causes of senile cataracts are associated with oxidative stress mechanisms. Glutatione peroxidase is selenoprotein, can reduce H2O2 and organic hydroperoxides into water and alcohol, glutatione peroxidase is found in high concentrations in the lens and plays an important role in protecting the lens by reducing the active oxygen species and peroxides such as disulfide and free radicals. Smoking is one factor that is thought to have a strong effect on the occurrence of cataracts caused by accumulation of metal ions in the tissues that directly compete with antioxidants and increase their inability to deposit in the lens. Objective: To know the difference of enzyme Glutathione Peroxidase lens level in cataract patients senile smoker and nonsmoker in connection with oxidative stress due to smoking risk factor. Method: Cross-sectional observational measurement study involving 58 subjects divided into 2 groups, Cataract of Senilis Smokers and Non- Smokers originating from RSUP H. Adam Malik Medan and network from October 2016 to Mai 2017. collected by eye lens after ECCE cataract surgery and SICS. Samples at the centrifuges and supernatants in homogenization and in the analysis of its GPX enzyme levels used spectrofotometry with a wavelength of 340 nm. Result: The enzyme glutatione peroxidase level of the lens in smokers cataracts was found to be lower than in the non-smoker cigerrete senile lenses in both range and average with a p value of less than 0,005
     
    Pendahuluan : Lensa mata sangat sensitif terhadap terjadinya stres oksidatif, sehingga banyak teori yang berkembang tentang penyebab katarak senilis yang berhubungan dengan mekanisme stres oksidatif. Glutatione perokside merupakan suatu Selenoprotein, dapat mereduksi H2O2 dan organic hydroperoxides menjadi air dan alkohol, Glutatione peroxidase dijumpai dalam konsentrasi yang tinggi pada lensa dan berperan penting dalam melindungi lensa dengan cara mengurangi spesies oksigen yang aktif dan peroksida seperti halnya disulfida dan radikal bebas. WHO memperkirakan 1,1 milyar orang mengkonsumsi rokok. Merokok adalah salah satu faktor yang diduga berpengaruh kuat terhadap terjadinya katarak yang disebabkan oleh akumulasi metal ion pada jaringan yang secara langsung bersaing dengan antioksidan dan meningkatkan ketidakmampuannya untuk berdeposit didalam lensa. Tujuan : Mengetahui perbedaan kadar enzim Glutation Peroxidase lensa pada penderita katarak senilis perokok dan bukan perokok sehubungan dengan adanya stres oksidatif karena faktor resiko merokok. Metode : Studi pengukuran secara observasional cross sectional melibatkan 58 subjek dibagi 2 kelompok, penderita Katarak Senilis Perokok dan Bukan Perokok yang berasal dari RSUP H. Adam Malik Medan dan jejaring dari bulan November 2016 hingga Mai 2017.yang dikumpulkan lensa matanya setelah operasi katarak ECCE dan SICS. Sampel di sentrifuse dan supernatannya di homogenisasi dan di analisa kadar aktifitas enzim GPX nya menggunakan spektrofotometri dengan panjang gelombang 340 nm. Hasil : Kadar Enzim Glutatione Peroxidase lensa penderita katarak senilis perokok secara signifikan lebih rendah dibandingkan kadar enzim Glutatione Peroxidase lensa katarak senilis bukan perokok baik secara kisaran maupun rerata dengan nilai p < 0,005.
