dc.description.abstractBackground: Increased cell population as result of proliferation increasing or decreased of death or differentiation cell. Tumors, mainly cancers which has higher proliferation activity, show prognostic factor and tumor aggressiveness. Annonaceous acetogenins(ACGs) which are the active agent of soursop leaves have a potential cytotoxic (antitumor) effect through inhibiting work on NADHubiqiunone oxidireductase (NADH-dehydrogenase or complex I). The purpose of this research to explore the impact of soursop leaves extract (anonna muricata) against tumor growth based on Ki-67 immunoexpression. Methods: This study used Wistar rats inoculated cancer, were divided into five groups C1, T1, C2, T2 and T3.T1 and T2 group was given soursop extract with dose of 2mg/day and T3 with dose of 4 mg/day. Meanwhile C1 and C2 group only feed by standart diet. The mass from rat is incised and prepared to slide histopathologic. Immunohistochemical with Ki-67 were performed. Result: There is significance expression within five groups with Kruskal-Wallis test p=0,02 (P<0,05). Mean–Whitnet test between C1 and T1 showed there is a significance difference expression with p=0,04 (p<0,05). There is also a significance difference Ki-67 expression in C2 and T2 group p=0,017 (P<0,05). Conclusion: Soursop leaves at 2 mg/day and 4 mg/day doses can be used to inhibit tumour growth. dc.description.abstractLatar Belakang: Meningkatnya jumlah sel dalam populasi tertentu dapat terjadi karena peningkatan proliferasi ataupun karena penurunan kematian atau diferensiasi sel. Proliferasi yang tinggi pada suatu tumor, khususnya kanker dapat menunjukkan prognosis dan agresifitas tumor. Bahan aktif daun sirsak yaitu Annonaceous acetogenins(ACGs) diteliti memiliki sifat sitotoksik (antitumor) akibat inhibisi kerja NADH-ubiqiunone oxidireductase (NADH-dehydrogenase atau complex I). Penelitian ini bertujuan untuk mengetahui pengaruh eskstrak daun sirsak terhadap pertumbuhan tumor yang dinilai berdasarkan imunoekspresi Ki-67. Metode: Penelitian ini menggunakan tikus Wistar yang telah diinokulasikan kanker, dan dibagi dalam 5 kelompok, yaitu C1, T1, C2, T2 dan T3. Kelompok T1dan T2 diberikan ekstrak daun sirsak 2 mg/hr, dan kelompok T3 dengan dosis 4 mg/hr. Sedangkan kelompok C1 dan C2 merupakan kelompok kontrol, hanya diberikan diet standar. Tumor yang tumbuh diambil, dibuat sediaan HE dan dipulas dengan pulasan imunohistokimia Ki-67. Hasil: Dengan uji Kruskal-Wallis didapati perbedaan bermakna antara kelima kelompok yaitu p=0,02 (P<0,05). Dilanjutkan dengan uji Mean-Whitney antara C1 dan T1 didapati perbedaan bermakna imunoekspresi dengan p=0,04 (P<0,05). Terdapat perbedaan bermakna imunoekspresi Ki-67 pada kelompok C2 dan T2 yaitu p=0,017 (P<0,05). Kesimpulan: Daun sirsak dapat menghambat pertumbuhan tumor dosis 2 mg/hari dan 4 mg/hari.
