    Identifikasi dan uji Antagonisme Jamur Endofit Tanaman Tebu (Saccharum Officinarum L.) terhadap Perkembangan Xanthomonas Albilineans L. dengan Metode Sterilisasi Autoklaf dan Membran Filter

    Abstract
    Siti Hardianti Wahyuni. 2015. Identification and Antagonism Test Endophytic Fungi of Sugarcane (Saccharum officinarum L.) to Growth Xanthomonas albilineans with Use Method Autoclaving and Membrane Filtration Sterilization. Supervised by Hasanuddin and Edison Purba. This research was conducted in a Laboratory of Agricuture Faculty and greenhouse of Agricuture Faculty, North Sumatra University, Medan, from April until December 2014. The research used completely randomized design with three factors and three replications. The first factor were endophytic fungi 1, endophytic fungi 2, endophytic fungi 3, endophytic fungi 4, endophytic fungi 5, endophytic fungi 6, endophytic fungi 7, and endophytic fungi 8. The second factor were dilution 10-1, 10-2, 10-3 dan 10-4. The three factor sterilization method were autoclaving and membrane filtration. This research aims to find potential endophytic fungi as biocontrol agents to against X. albilineans bacterial vascular wilt on sugarcane used method autoclaving and membrane filtration sterilization. The results showed eight the endophytic fungi were endophytic fungi 1 (Aspergillus sp.), endophytic fungi 2 (Aspergillus sp.), endophytic fungi 3 (Penicillium sp.), endophytic fungi 4 (Penicillium sp.), endophytic fungi 5 (Cephalosporium sp.), endophytic fungi 6 (Curvularia sp.), endophytic fungi 7 (Fusarium sp.), dan endophytic fungi 8 (Hormiscium sp.) on sugarcane variety BZ 134. Endophytic fungi can control bacterial vascular wilt X. albilineans on sugarcane. Endophytic fungi 3 (Penicillium sp.), endophytic fungi 5 (Cephalosporium sp.) and endophytic fungi 8 (Hormiscium sp.) were more endure to autoclaving sterilization than other endophytic fungi. The best diameter of inhibiting zone on Penicillium sp. 11,82 mm with use autoclaving sterilization. The incidence and severity of disease is highest in treatment interaction endophytic fungi 8 (Hormiscium sp.) with membrane filtration sterilization and dilution 104 were 15,87 % dan 35,18 %.

Penelitian ini dilaksanakan di Laboratorium Penyakit Tumbuhan dan Rumah Kaca Fakultas Pertanian Universitas Sumatera Utara, Medan pada bulan April sampai Desember 2014. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) Faktorial dengan tiga faktor dan tiga ulangan. Faktor pertama jamur endofit (J) yaitu jamur endofit 1, jamur endofit 2, jamur endofit 3, jamur endofit 4, jamur endofit 5, jamur endofit 6, jamur endofit 7, dan jamur endofit 8. Faktor kedua pengenceran (P) yaitu 10-1, 10-2, 10-3 dan 10-4. Faktor ketiga metode sterilisasi yaitu autoklaf dan membran filter. Penelitian ini bertujuan untuk menguji potensi jamur endofit terhadap X. albilineans penyebab penyakit vaskular bakteri pada tanaman tebu dengan menggunakan dua metode sterilisasi autolaf dan membran filter. Hasil penelitian menunjukkan terdapat 8 jenis jamur endofit yaitu jamur endofit 1 (Aspergillus sp.), jamur endofit 2 (Aspergillus sp.), jamur endofit 3 (Penicillium sp.), jamur endofit 4 (Penicillium sp.), jamur endofit 5 (Cephalosporium sp.), jamur endofit 6 (Curvularia sp.), jamur endofit 7 (Fusarium sp.), dan jamur endofit 8 (Hormiscium sp.) pada tanaman tebu varietas BZ 134. Jamur endofit dapat menghambat bakteri penyebab penyakit vaskular bakteri X. albilineans pada tanaman tebu. Jamur endofit 3 (Penicillium sp.), jamur endofit 5 (Cephalosporium sp.) dan jamur endofit 8 (Hormiscium sp.) merupakan jamur yang lebih tahan disterilisasi autoklaf dibandingkan jamur endofit yang lain. Diameter zona hambat terbaik didapat pada Penicillium sp. dengan diameter zona hambat 11,82 mm menggunakan sterilisasi autoklaf. Kejadian dan keparahan penyakit tertinggi terdapat pada perlakuan interaksi jamur endofit 8 (Hormiscium sp.) menggunakan sterilisasi membran filter dan pengenceran 104 yaitu sebesar 15,87 % dan 35,18 %.
