    Hubungan Ekspresi Pulasan Imunohistokimia Protein Gene Product (PGP9.5) pada Lapisan Fungsional Endometrium dan Jaringan Endometriosis di Ovarium

    Abstract
    Background. Endometriosis is a condition characterized by epithelial endometrial gland and stroma found outside of the uterine cavity. More recently, there is accumulating literature describing nerve fiber density differences between women with diagnosed endometriosis and women without endometriosis. Neuronal markers were used to discover nerve fibers presence within endometrial layers such as protein gene product 9.5 (PGP9.5) which was a specific marker for unmyelinated and myelinated nerve fibers. PGP9.5 has also been demonstrated in various endometriotic lesions such as ovarian endometriotic lesions. Objectives. To investigate the expression of PGP9.5 in nerve fibers within functional layer of endometrium and ovarian endometriotic lesions and to analyze their correlations. Methods. This is an analytical descriptive study with cross sectional approach. The samples are the ovarian endometriotic lesions and the endometrial layer tissue embbeded in paraffin wax. Fifty paraffin embbeded tissues will be stain with PGP9.5. The expressions and their correlations will be analyzed with statistical analysis. Results. Nerve fibres stained with PGP9.5 were detected in functional layer of endometrium in 16% of women with ovarian endometriotic lesions and the density was 0.049±0.147/mm2. Nerve fibres stained with PGP9.5 were detected in ovarian endometriotic lesions in 12% of women with ovarian endometriotic lesions and the density was 0.173±0.498/mm2. No correlations found between PGP9.5-immunoactive fibres expression in functional layer of endometrium and in ovarian endometriotic lesions (p=0.422). Conclusions: No correlations found between PGP9.5-immunoactive fibres expression in functional layer of endometrium and in ovarian endometriotic lesions.
     
    Latar Belakang. Endometriosis adalah kondisi dimana jaringan endometrium yang terdiri dari epitel kelenjar maupun stroma ditemukan pada lokasi di luar rongga uterus. Pada penelitian endometriosis terbaru saat ini didapati bahwa terdapat perbedaan kepadatan distribusi serabut saraf antara perempuan penderita endometriosis dengan perempuan tanpa endometriosis. Beberapa penanda saraf digunakan untuk mengidentifikasi serabut saraf pada lapisan endometrium seperti Protein Gene Product (PGP9.5) yang merupakan penanda spesifik untuk serabut saraf bermyelin dan tidak bermyelin. Ekspresi PGP 9.5 dapat dijumpai pada berbagai lesi endometriosis termasuk lesi endometriosis di ovarium. Tujuan. Tujuan penelitian ini adalah melihat apakah serabut saraf yang dipulas dengan PGP9.5 dijumpai pada lapisan fungsional endometrium dan jaringan endometriosis di ovarium pada penderita endometriosis ovarium dan melihat hubungan ekspresi keduanya. Metode. Penelitian ini adalah penelitian deskriptif analitik dengan pendekatan cross sectional. Sampel penelitian adalah blok parafin berasal dari jaringan endometriosis di ovarium dan jaringan endometrium uterus. Sejumlah 50 sampel dipulas dengan imunohistokimia PGP9.5 dan hasil ekspresi pulasan serta hubungannya dianalisis dengan uji statistik. Hasil. Serabut saraf yang terpulas PGP9.5 dijumpai pada lapisan fungsional endometrium pada 16% penderita dan distribusinya adalah 0.049±0.147/mm2. Serabut saraf yang terpulas PGP9.5 pada lesi endometriosis ovarium dijumpai pada 12% penderita dan distribusinya adalah 0.173±0.498/mm2. Tidak terdapat hubungan antara serabut saraf yang terpulas dengan PGP9.5 pada lapisan fungsional endometrium dan pada lesi endometriosis di ovarium (p=0.422). Kesimpulan. Tidak terdapat hubungan antara serabut saraf yang terpulas dengan PGP9.5 pada lapisan endometrium dan pada lesi endometriosis di ovarium.
