dc.description.abstractThe aim of this research was to study the effect supplementation of Duck egg yolk in two-steptm-extender on motility, viability, and morfology sperm of limousin bull. The research used randomized completely design with five repetitions from five different bulls. Sperm was stored for ten days. Sperm motility was observed using light microscopy (400x) at temperature of 37°C. Sperm viability was observed with eosin staining by light microscopy (400x). Sperm morfology was observed with metilen blue staining by light microscopy (400x). The result of the research showed that egg yolk supplementation in two-steptm extender affected the motility and viability, morfology sperm of limousin bull during storage at 4- 5°C. The best egg yolk concentration was 15% in protecting the motility (41,00±2,24%) viability (74.44±4,77%) and morfology sperm (69,91±1,98%) of Limousin bull after storage at 4-5°C.en_US
dc.description.abstractTujuan dari Penelitian ini adalah mengetahui pengaruh penambahan kuning telur bebek dalam pengencer two-step Tm extender terhadap motilitas, viabilitas dan morfologi spermatozoa sapi limousin. Penelitian menggunakan eksperimen laboratorium menggunakan pola rangcangan acak lengkap (RAL) dengan 5 kali ulangan dari 5 ejakulasi dari sapi yang sama. Spermatozoa disimpan selama sepuluh hari. Motilitas spermatozoa diamati dengan mikroskop cahaya (200x) pada suhu 370C. Viabiltas spermatozoa diamati dengan pewarnaan eosin menggunakan mikroskop cahaya (400x), morfologi spermatozoa diamati dengan pewarnaan Metilen blue menggunakan mikroskop cahaya (400x). Hasil penelitian menunjukkan suplementasi kuning telur bebek pada pengencer two-step Tm extender berpengaruh terhadap motilitas dan viabilitas, morfologi spermatozoa sapi limousin selama penyimpanan pada suhu 4-5° C. Konsentrasi kuning telur terbaik adalah 15% dalam mempertahankan motilitas (41,00%±2,24) viabilitas (74,44±4,77%) dan morfologi (69,91±1,98%) spermatozoa sapi Limousin setelah penyimpanan empat Hari pada suhu 4-50 c.en_US
