Potential of entomopathogenic nematodes Steinernema sp. of the coffee berry
borer, Hypothenemus hampei in North Sumatra has not been reported. For it has
done mortality test local isolate Steinernema sp. against H. hampei in the
laboratory and field. The study aimed to determine the concentrations of
Steinernema sp. local isolates against H. hampei in the laboratory and field. The
study was conducted in laboratory Balai Besar Perbenihan and Proteksi
Tanaman (BBPPTP) Medan and coffee plantation in Subdistrict Habinsaran
District Toba Samosir, North Sumatra. The treatments were tested in laboratory
is the density of nematodes with various concentrations of infective juveniles
4500, 5000, 5500, 6000 ji/ml of sterile water and control (without treatment). The

the field, the highest mortality of larvae, pupa and imago H. hampei is in 6500
ji/ml of sterile water respectively 33.13, 33.29 and 39.53%. H. hampei mortality
increases with the density of nematodes. Potensi nematoda entomopatogen Steinernema sp. terhadap penggerek
buah kopi, Hypothenemus hampei di Sumatera Utara belum pernah dilaporkan.
Untuk itu telah dilakukan uji mortalitas Steinernema sp. isolat lokal terhadap
H. hampei di laboratorium dan lapangan. Penelitian bertujuan untuk mendapatkan
konsentrasi Steinernema sp. isolat lokal yang tepat terhadap H. hampei di
laboratorium dan lapangan. Penelitian dilakukan di laboratorium lapangan Balai
Besar Perbenihan dan Proteksi Tanaman Perkebunan (BBPPTP) Medan dan
kebun kopi petani di Kecamatan Habinsaran Kebupaten Toba Samosir Provinsi
Sumatera Utara. Perlakuan yang dicobakan dalam penelitian di laboratorium
adalah kerapatan nematoda terhadap larva, pupa dan imago H. hampei dengan
berbagai konsentrasi yaitu 4500, 5000, 5500, 6000 juvenil infektif (ji)/ml air steril
dan kontrol (tanpa perlakuan). Perlakuan yang dicobakan di lapangan adalah
kerapatan nematoda dengan tingkat konsentrasi 5000, 5500, 6000, 6500 ji/ml air
steril dan kontrol (tanpa perlakuan). Aplikasi dilakukan dengan menyemprotkan
suspensi ke buah-buah kopi matang yang terserang H. hampei. Perbanyakan
nematoda dilakukan secara in vivo menggunakan ulat hongkong (Tenebrio
molitor). Hasil penelitian menunjukkan pada pengujian di laboratorium mortalitas
larva H. hampei tertinggi terdapat pada perlakuan 5000 ji/ml air steril sebesar
37,58, pupa 35,69 dan imago 32,55%. Mortalitas larva, pupa dan imago H.
hampei di lapangan tertinggi pada perlakuan 6000 ji/ml air steril masing-masing
sebesar 33,13, 33,29 dan 39,53 %. Mortalitas H. hampei meningkat seiring
dengan bertambahnya kerapatan nematoda.
