dc.description.abstractThe use of microorganisms as biocontrol agents have been known to reduce soil pollution due to the use of chemical compounds. Among the microorganisms chitinolytic bacteria have been utilized as biocontrol agents to inhibit both fungal pathogens on plants and animals. In this study, chitinolytic bacteria isolated from shrimp waste from shrimp processing plant and shrimp waste disposal. Among Fiftyfive bacteria isolated, ten bacteria were found have chitinolytic activity in chitin medium with chitinolytic index up to 2.8. Ten isolates bacteria were tested for in vitro antagonism against several fungal pathogens. Two of ten isolates i.e NR09 and NR02 were selected as the best isolates. For further study, NR09 showed potential to inhibit Rigidoporus microsporus, Sclerotium rolfsii and Fusarium oxysporum by 63.6%, 64.6% and 75,6% respectively. While NR02 showed potential to inhibit Curvularia sp., Saprolegnia sp. and C. albicans by 68.9%, 54.2% and 52.3% respectively. Microscopic observation antifungal effect on fungal pathogen revealed swolen, lytic and twisted hyphae. The free cell-extract of isolate NR09 was capable to inhibit the growth of fungal pathogen in 28% - 65.5%.en_US
dc.description.abstractPenggunaan mikroorganisme sebagai agen biokontrol telah banyak digunakan mengingat perannya dalam mengurangi pencemaran tanah akibat penggunaan senyawa kimia. Diantara mikroorganisme yang digunakan adalah bakteri kitinolitik sebagai agen biokontrol untuk mengatasi patogen baik pada tanaman maupun hewan. Pada kajian ini bakteri kitinolitik diisolasi dari limbah udang yang berasal dari pabrik pengolahan udang dan tanah tempat pembuangan limbah udang. Dari 55 isolat bakteri yang diperoleh, didapatkan 10 isolat bakteri kitinolitik potensial dengan indeks kitinolitik mencapai 2,8. Uji antagonis isolat bakteri potensial terhadap beberapa jamur patogen secara in vitro menunjukkan bahwa isolat bakteri NR09 memiliki persentase penghambatan yang tertinggi terhadap Rigidoporus microsporus, Sclerotium rolfsii dan Fusarium oxysporum dengan persentasi penghambatan masingmasing 63,6%. 64,6% dan 75,6%. Penghambatan tertinggi terhadap Curvularia sp., Saprolegnia sp dan C. albicans oleh isolat bakteri NR02 dengan persentase penghambatan masing-masing 68,9%, 54,2% dan 52,3%. Pengamatan mikroskopis hifa abnormal pada jamur patogen menunjukkan adanya hifa yang menggulung, lisis dan membengkok. Ekstrak-free cell isolat bakteri NR09 mampu menghambat pertumbuhan jamur patogen dengan persentase penghambatan antara 28% - 65,5%.en_US
