dc.description.abstract | Picria felterrae Lour (Schropulareaceae) is widely used in traditional
medicine and reported to have activities as analgetic, antiinflamatory, antidiabetic,
and antioxidant. The objective of this study was to evaluate the activies of nhexane
fraction (EnHPT), ethyl acetate fraction (EEAPT) and ethanol fraction of
puguntano leaf (EEPT) on rat’s cardiovascular system.
Puguntano was collected from Sidikalang, Dairi region, North Sumatera.
The herbarium specimen of the plant was deposited at the Lembaga Ilmu
Pengetahuan Indonesia (LIPI). Fractions of EnHPT, EEAPT and EEPT were
prepared by remaceration. Phytochemical screening was carried out on the dried
plants material and its fractions. Diuretic activity of each fraction was examined
in rats by measuring urine volume at interval of one hour at 1, 2, 3, 4, 5 and 6
hours after the administration of fraction. Control group received 0.5% of natrium
carboxymethyl cellulose (Na CMC) and positive control group received
furosemide (10 mg/kg bw/day). Varied doses of EnHPT, EEAPT and EEPT (100,
200, 400 and 800 mg/kg bw/day) were orally administered to the test groups . The
total volume of urine excreted was expressed in ml/6 hr. Diuretic index values of
the test groups were calculated by comparing with control and furosemide groups,
respectively. Acute toxicity was assessed according to BPOM guidline.
Hypertensive activity was examined in normotensive group, 2.5% natrium
chloride-induced group, and L-Name (Nω-L-arginine methyl ester)-induced group
by non-invasive tail-cuff method. In normotensive group, EnHPT, EEAPT and
EEPT doses of 400 and 800 mg/kg bw/day were examined for its hipotensive
effect. In NaCl group, 2.5% NaCl solution and methylprednisolon dose of 1,5
mg/kg bw/day was administered orally in rats for 14 days to induce hypertension.
Bisoprolol (0.0017 mg/kg bw/day; p.o) used as positive control. Antihypertensive
effect of EnHPT, EEAPT, and EEPT at doses of 400 and 800 mg/kg bw/day were
examined for 7 days. In L-Name group, L-Name (75 mg/kg bw/day) was
administered orally in rats for 14 days to induce hypertension and then
antihypertensive effect of EnHPT, EEAPT, and EEPT doses 400 and 800 mg/kg
bw/day were examined for 7 days. Cardiotonic activity of the plant fraction was
examined on isolated rat heart preparation using hypodynamic Krebs-Henseleit
(KH) solution on Langendorff’s assembly.
The phytochemical analysis showed the presence of active phytochemical
groups such as triterpenoid, steroid, glycoside, flavonoid, tannin and saponin in
simplicia; steroid and triterpenoid in EnHPT; flavonoid, glycoside, tannin and
saponin in EEAPT; glycoside and saponin in EEPT. Plant fractions demonstrated
significant diuretic activity (p < 0.05), and kaliuretic (p < 0.05) effects in treated
groups in a dose-dependent manner. Diuretic index showed good diuretic activity
of the EnHPT at the dose of 800 mg/kg compared with that of the control and
furosemide. No lethal effects were observed among mice even at the high dose of 5000 mg/kg. Histophatological examination showed that EnHPT, EEAPT and
EEPT 5000 mg/kgBB caused degeneration hydrofic and necrosis.
Antihypertensive activity showed that EEPT did not lower the blood pressure
(P>0.05) but EnHPT and EEAPT decreased the systolic blood pressure (SBP),
diastolic blood pressure (DBP) and mean arterial pressure (MAP) in normotensive
group (P<0.05), respectively. EnHPT, EEAPT and EEPT decreased SBP, DBP,
MAP in NaCl-induced hypertensive group without affecting the heart rate
(HR)(P>0.05). Bisoprolol was effective to decrease the Wistar rats blood pressure
(P<0.05). EnHPT, EEAPT and EEPT decreased SBP, DBP, MAP in L-Nameinduced
hypertensive model without affecting HR (P<0.05). The positive cardiac
inotropic and chronotropic effects were showed by EnHPT. EEAPT 0.5 mg
showed positive inotropic effects compared to KH and digoxin without affecting
HR (P<0.05). EEPT has no inotropic and chronotropic effects.
It was concluded that EnHPT at the dose of 800 mg/kg significantly
increased the urinary volume and concentration of urinary electrolytes with no
signs of toxicity and therefore, is a potential diuretic. All extract showed
antihypertensive effect in NaCl and L-Name-induced hypertensive rats. EEAPT
has positive inotropic and chronotropic effect. | en_US |
dc.description.abstract | Picria felterrae Lour (Schropulariaceae) telah digunakan dalam pengobatan
tradisional dan memiliki aktivitas sebagai analgetik, antiinflamasi, antidiabetik
dan antioksidan. Tujuan penelitian ini adalah untuk mengevaluasi aktivitas fraksi
n-heksan (EnHPT), etilasetat (EEAPT) dan etanol (EEPT) daun puguntano pada
sistem kardiovaskular tikus.
Puguntano dikumpulkan dari Sidikalang, Kabupaten Dairi, Sumatera Utara.
Simplisia tumbuhan diidentifikasi oleh LIPI. Fraksi EnHPT, EEAPT dan EEPT
diperoleh menggunakan metode remaserasi. Skrining fitokimia dilakukan
terhadap simplisia dan fraksi tumbuhan. Aktivitas diuretik dilakukan pada tikus
dengan mengukur volume urin pada jam ke 1, 2, 3, 4, 5 dan 6 setelah pemberian
fraksi. Kelompok kontrol diberi Na CMC, kontrol positif diberi furosemide (10
mg/kgbb/hari) dan kelompok uji diberi fraksi dosis 100, 200, 400 dan 800
mg/kgbb/hari secara oral. Urin dikumpulkan dan volume urin total dinyatakan
dalam ml/6 jam. Nilai indeks diuretik ditentukan berdasarkan perbandingan
volume urin kelompok perlakuan terhadap kelompok kontrol dan furosemid.
Toksisitas akut ditentukan berdasarkan pedoman BPOM. Aktivitas hipertensi
dilakukan pada tikus normotensi dan dua model tikus hipertensi: yang diinduksi
NaCl 2,5% dan yang diinduksi L-Name (Nω-L-arginin metil ester) menggunakan
metode non-invasive tail-cuff. Pada kelompok normotensi, EnHPT, EEAPT dan
EEPT dosis 400 dan 800 mg/kg bb/hari diberikan untuk mengetahui efek
hipotensi. Pada kelompok tikus hipertensi NaCl, tikus diinduksi dengan larutan
NaCl 2,5% dan metilprednisolon 1,5 mg/kg bb/hari secara oral selama 14 hari.
Bisoprolol digunakan sebagai pembanding. EnHPT, EEAPT, dan EEPT dosis 400
dan 800 mg/kg bb/hari diberikan untuk mengetahui efek antihipertensinya selama
7 hari. Pada kelompok L-Name, L-Name (75 mg/kg bb/hari) diberikan ke tikus
secara oral selama 14 hari dan kemudian EnHPT, EEAPT, EEPT dosis 400 dan
800 mg/kg bb/hari diberikan untuk mengetahui efek antihipertensinya selama 7
hari. Aktivitas kardiotonik fraksi dilakukan pada isolat jantung tikus yang
disiapkan dalam larutan fisiologi krebs-Henseleit (KH) menggunakan alat
langendorf.
Analisis fitokimia menunjukkan adanya senyawa aktif dalam tiap fraksi
seperti triterpenoid, steroid, flvonoid, glikosida, tanin dan saponin pada simplisia,
triterpenoid dan steroid pada EnHPT, glikosida, flavonoid, tanin dan saponin pada
EEAPT, glikosida dan tanin pada EEPT. Hasil penelitian menunjukkan bahwa
terjadi peningkatan efek diuretik dan kaliuretik pada kelompok perlakuan secara
signifikan (P<0,05). Indek diuretik menunjukkan aktivitas diuretik EnHPT 800
mg/kgbb yang paling baik dibandingkan dengan kontrol dan furosemid. Efek letal
tidak terjadi diantara mencit hingga pemberian dosis tertinggi yaitu 5000
mg/kgbb. Hasil histopatologi menunjukkan bahwa EnHPT, EEAPT dan EEPT
5000 mg/kgBB menyebabkan degenerasi hidrofik dan nekrosis pada ginjal dan hati. Aktivitas antihipertensi menunjukkan bahwa EEPT tidak menurunkan
tekanan darah (P>0,05) tetapi EnHPT dan EEAPT menurunkan tekanan darah
sistol (TDS), tekanan darah diastol (TDD), dan rerata tekanan arteri (TAR) pada
kelompok tikus normotensi (P<0,05). EnHPT, EEAPT dan EEPT menurunkan
TDS, TDD, TAR kelompok tikus hipertensi yang diinduksi NaCl tanpa
menpengaruhi denyut jantung (DJ) (P>0,05). Bisoprolol efektif menurunkan
tekanan darat tikus Wistar (P<0,05). EnHPT, EEAPT dan EEPT menurunkan
TDS, TDD, TAR kelompok tikus hipertensi yang diinduksi L-Name tanpa
menpengaruhi DJ (P>0,05). Efek inotropik dan kronotropik positif ditunjukkan
oleh EnHPT. EEAPT 0.5 mg menunjukkan inotropik positif dibandingkan KH
dan digoksin tanpa mempengaruhi DJ. EEPT tidak memiliki efek inotropik dan
kronotropik.
Kesimpulan penelitian ini menunjukkan bahwa EnHPT dosis 800 mg/kgbb
meningkatkan volume urin dan elektrolit secara signifikan tanpa menunjukkan
gejala toksik sehingga berpotensi sebagai diuretik. Semua fraksi menunjukkan
aktivitas antihipertensi pada tikus yang diinduksi NaCl dan diinduksi L-Name.
EEAPT mempunyai efek inotropik dan kronotropik positif. | en_US |
