dc.description.abstract | Minimnya ketersediaan benih sumber kentang bersertifikat dan
berkualitas, serta terhambatnya diseminasi dan adopsi varietas kentang unggul
menjadi salah satu faktor krusial rendahnya produktivitas kentang nasional. Salah
satu solusi yang dapat ditempuh yakni dengan pemanfaatan umbi mikro sebagai
propagul yang lebih efisien. Oleh karenanya, penelitian ini bertujuan untuk
memperoleh sistem kultur yang tepat dalam budidaya umbi mikro secara in vitro,
dengan menganalisis pengaruh dan interaksi aplikasi dua sistem kultur berbeda,
yaitu kultur konvensioanal dan Temporary Immersion System (TIS) Bioreactor
terhadap sejumlah varietas kentang berbeda. Penelitian ini menggunakan
Rancangan Acak Lengkap (RAL) 2 faktor. Faktor pertama berupa perlakuan jenis
sistem kultur (konvensional dan TIS Bioreactor). Faktor kedua berupa varietas
kentang (Atlantic Malang, Dayang Sumbi Agrihorti, Granola L., dan Maglia).
Hasil penelitian menunjukkan bahwa pada tahapan multipikasi tunas dan produksi
umbi mikro kultur TIS Bioreactor memiliki rata-rata tertinggi pada semua
parameter penelitian, dibandingkan kultur konvensional. Adapun terkait varietas,
Dayang Sumbi unggul dalam jumlah tunas, akar primer, diameter, bobot basah
dan waktu terbentuknya umbi, Granola L. unggul dalam tinggi planlet serta
jumlah umbi. Sementara itu, Atlantic Malang unggul dalam laju multipikasi dan
jumlah nodus, sedangkan Maglia unggul dalam jumlah daun. | en_US |
dc.description.abstract | As the national potatoes growth has become lower from year to year, it is
now known that the problems came from the lack of certified potato seed varieties
and the minimum access to a sophisticated technology to make a good potato
variety. The solution that can be made is to utilize microtubers as an efficient
factors. The purpose of this research was to find the most effective method in
micro tubers cultivation in vitro by comparing the conventional tissue culture
method and the Temporary Immersion System (TIS) Bioreactor method to four
different potato varieties (Granola L., Dayang Sumbi, Atlantic Malang, Maglia).
This research used a split plot design with a completely randomize design by
using two factorials. The result of this study showed that the microtubers in
multipication and production step using the TIS Bioreactor method had higher
average of parameters than the conventional tissue culture method. As the variety
details, Dayang Sumbi had the highest parameter such as the number of sprouts,
primer roots, diameter, fresh weights, and time growth. Granola L. excelled in
planlet height and the number of tubers. Meanwhile, Atlantic Malang excelled in
the multiplication rate and the number of nodes, and then Maglia excelled in the
