dc.description.abstract | One plant that has medicinal properties is the petai cina (Leucaena leucocephala (Lam.) de Wit) including the Leguminosea/Fabaceae family, which used as an antidiarrheal, anthelmintic, and antiseptic. This study on the standardization of ethanol extract and the isolation of flavonoid compounds from petai cina seeds was carried out as an effort to ensure that the final product had certain parameter values so as to produce extracts of good quality. The aim of this research is to determine specific and nonspecific standard parameter data to determine specific and non-specific standard parameter data as well as to determine the acute toxicity potential of the ethanol extract of petai cina seeds (EEBPC) and also to find out the flavonoids in petai cina seeds.
The simplicia powder of petai cina seeds was macerated with 80% ethanol. The extract obtained was fractionated using n-hexane and ethyl acetate solvents. Standardization was carried out on specific and nonspecific parameters, determination of acute toxicity testing that referred to OECD (Organization of Economic Cooperation and Development) 420. Isolation of flavonoid compounds from ethyl acetate fraction, was carried out by paper chromatography and preparative paper chromatography using the BAW mobile phase (n-butanol: acetic acid: water = 4:1:5), HCl 1% and acetic acid 15%. Isolate compound were characterized by spectrophotometry ultraviolet (UV) using the shifts reagent method.
The results of the specific standardization of the ethanol extract of petai cina seeds were organoleptically obtained by a thick green-blackish extract with a bitter taste and distinctive smell, a water-soluble extract content of 10.55%, an ethanol soluble extract content 1,68%. The results of non-specific standardization of the ethanol extract of petai cina seeds obtained moisture content of 5.33%, total ash content of 6.23%, acid insoluble ash content of 0.77%, specific gravity of 0.8172, Pb metal contamination of 0 ppm, metal contamination Cd 0 ppm, no bacterial contamination and mold/yeast contamination. The results of the EEBPC acute toxicity test showed no toxic symptoms until the 14th day, the average body weight results for each group after being given EEBPC also did not show a significant difference between the control group and treatment where (p > 0.05). The isolates obtained were yellowish green (Rf = 0.87), namely the flavanone group with a maximum wavelength of 330 nm (band I) and 275 nm (band II). | en_US |
