Abstract

Poguntano (Picria fel-terrae Lour.) Is a medicinal plant in North Sumatra which has been used in traditional medicine to treat various diseases. Poguntano herbal extract contains flavonoids, alkaloids, glycosides, saponins, tannins and steroids/triterpenoids which have the potential to be developed into immunomodulators. This study aimed to compare the immunostimulant activity of poguntano herb n-hexane extract (PHNHE), poguntano herb ethyl acetate extract (PHEAE) and poguntano herb ethanol extract (PHEE) in immunosuppression rats that have been infected Staphylococcus aureus. Poguntano herb were dried, ground, then extracted by maceration method using n-hexane, ethyl acetate and ethanol as solvents. The test animals used were male rats that were given the extract on the first day to the 14th day, on the 4th day the test animals were infection by Staphylococcus aureus bacteria then induced cyclophosphamide 70 mg / kg bw on day 8, and 13 then tested for immunostimulant activity using the total analysis method. leukocytes, differential leukocytes, Ig G antibody assay and expression of cytokines IFN-, IL-2, IL-4 by ELISA method. Poguntano (Picriafel-terrae Lour.) merupakan salah satu tanaman obat di Sumatera Utara yang telah digunakan dalam pengobatan tradisional untuk mengatasi berbagai penyakit.Ekstrak herba poguntano mengandung senyawa flavonoid, alkaloid, glikosida, saponin, tanin dan steroid/triterpenoid yang berpotensi dikembangkan menjadi imunomodulator. Penelitian ini bertujuan untuk menentukanaktivitas imunostimulan ekstrak n-heksana herba poguntano (ENHHP), ekstrak etilasetat herba poguntano (EEAHP) dan ekstrak etanol herba poguntano (EEHP) pada tikus imunosupresiyang telahdiinfeksi Staphylococcus aureus. Ekstrak diperoleh secara maserasi bertingkat dengan menggunakan pelarut n-heksana, etilas etat dan etanol. Hewan uji yang digunakan adalah tikus jantan yang diberikan ekstrak pada hari pertama sampai hari ke 14, pada hari ke 4 hewan uji diinfeksi bakteri Staphylococcus aureus selanjutnya diinduksi siklofosfamid 70 mg/kg bb pada hari ke 8 dan 13 selanjutnya diuji aktivitas imunostimulan dengan metode analisis total leukosit, diferensial leukosit, uji antibodi Ig G dan ekspresisitokin IFN , IL-2, IL-4 dengan metode ELISA. Hasil pengujian total leukosit dan diferensial leukosit pada ENHHP dan EEAHP dosis 50 mg/kg bb tidak dapat meningkatkan jumlah total leukosit dan diferensial leukosit, pada EEAHP dosis 100, 200 mg/kg bb dan EEHP dosis 50, 100, 200 mg/kg bb dapat meningkatkan total leukosit dan diferensial leukosit dengan perberbedaan yang signifikan terhadap kelompok negatif (p<0,05) dan tidak berbeda signifikan dengan levamisol dan kelompok normal (p>0,05). Hasil pengujian level Ig-G pada ENHHP dan EEAHP dosis 50 mg/kg bb tidak dapat meningkatkan level Ig-G, pada EEAHP dosis 100, 200 mg/kg bb dan EEHP dosis 50, 100, 200 mg/kg bb dapat meningkatkan level Ig-Gdengan perberbedaan yang signifikan terhadap kelompok negatif (p<0,05) dan tidak berbeda signifikan dengan levamisol dan kelompok normal (p>0,05). Hasil pengujian level sitokin IL-2, IL-4 dan IFN- pada ENHHP, EEAHP dosis 50 mg/kg bb dan EEHP dosis 50 mg/kg bb tidak dapat meningkatkan level sitokin IL-2, IL-4 danIFN-, padaEEAHP dosis 100, 200 mg/kg bb dan EEHP dosis 100, 200 mg/kg bb dapat meningkatkan level sitokin IL-2, IL-4 dan IFN- dengan perberbedaan yang signifikan terhadap kelompok negatif (p<0,05) dan tidak berbeda signifikan dengan levamisol dan kelompok normal (p>0,05). Berdasarkan hasil penelitian inimenunjukkan EEAHP dan EEHP dapat meningkatkan total leukosit, diferensial leukosit, antibodi Ig-G, ekspresi sitokin IL-2, IL-4 danIFN-, sedangkan ENHHP tidak dapat meningkatkan total leukosit, diferensial leukosit, titer antibodi dan ekspresi sitokin, sehingga EEAHP dan EEHP efektif sebagai aktivitas imunostimulan.
