    Pengaruh Ekstrak Etanol Daun Mobe (Artocarpus lakoocha Buch. Ham) Terhadap Biofilm dan Kebocoran Membran Sel Streptococcus mutans

    Abstract
    Background: Fixed orthodontic appliances can increase biofilm and Streptococcus mutans. Streptococcus mutans is a cariogenic oral bacteria that causes caries. Many natural ingredients extracted to reduce the biofilm and the number of cariogenic oral bacteria in the oral cavity. One of the potential natural ingredient is mobe plant. Mobe plants contain active antibacterial compounds artocarpin, oxyresveratrol, phenols, flavonoids, and tannins. The purpose of the study: to determine the antibacterial activity, antibiofilm, and leakage of DNA and protein from the mobe plant (Artocarpus lakoocha Buch. Ham). Methods: The antibacterial activity was evaluated using the diffusion method to determine the MIC. Antibiofilm activity and DNA and protein leakage were measured using spectrophotometry. Results: This study showed the ethanolic extract of mobe leaves had antibacterial and antibiofilm effects and cause DNA leakage with an MIC of 3.125 mg/ml. The ethanolic extract of mobe leaves with a concentration of 25 mg/ml can cause protein leakage (abs 280 nm). The ethanolic extract of mobe leaves 3.125 mg/ml and 0.1% chlorhexidine have the same effectiveness against Streptococcus mutans in protein leakage test (abs 280 nm). Conclusion: The ethanolic extract of mobe leaves can be able to inhibit the growth of Streptococcus mutans with a minimum inhibition of 3.125% concentration.
     
    Latar belakang: Pemakaian alat ortodonti cekat dapat meningkatkan biofilm dan bakteri Streptococcus mutans. Streptococcus mutans adalah bakteri penyebab karies yang kariogenik. Banyak bahan alami yang diekstrak untuk menurunkan biofilm dan jumlah bakteri kariogenik pada rongga mulut. Salah satu bahan alami yang berpotensi adalah tumbuhan mobe. Tumbuhan mobe memiliki senyawa antibakteri yang aktif, yaitu: artocarpin, oxyresveratrol, fenol, flavonoid, dan tanin. Tujuan penelitian: untuk mengetahui aktivitas antibakteri, antibiofilm, dan kebocoran DNA dan protein dari tumbuhan mobe (Artocarpus lakoocha Buch. Ham). Metode penelitian: Aktivitas antibakteri dievaluasi menggunakan dengan metode difusi untuk menentukan MIC. Aktivitas antibiofilm dan kebocoran DNA dan protein diukur menggunakan dengan Spektofotometri. Hasil: Penelitian ini menunjukkan ekstrak etanol daun mobe memiliki efek antibakteri dan antibiofilm dengan MIC 3,125 mg/ ml. Ekstrak etanol daun mobe 3,125 mg/ ml dapat menyebabkan kebocoran DNA dengan Hasil kebocoran DNA dan protein pada ekstrak mobe 3,125 mg/ml tidak berbeda signifikan dengan kloroheksidin 0,1%. Kesimpulan: Ekstrak daun mobe mampu menghambat pertumbuhan Streptococcus mutans dengan daya hambat minimal adalah konsentrasi 3,125%.
