Dental caries and periodontal disease might synergistically and negatively associated, or completely independent of each other. Positive association may be related to the microbiological etiology and biofilm formation. The cause of periodontal disease which is one of the risk factors of orthodontic treatment is Staphylococcus aureus. When the demineralization occurs, mouthwash is needed to assist the remineralization. Various studies have reported the activity of antibacterial and antibiofilm from natural ingredients, one of them is butterfly pea (Clitoria ternatea L.) as an antimicrobial. The aim of this research is to determine the activity of butterfly pea extract of the antibacterial, antibiofilm, leakage of DNA, protein, calcium and potassium metals against S. aureus. The method of this research is an experimental laboratory with Post Test Only Group Design. The sample of this study is the ethanol extract of butterfly pea which was diluted using DMSO to a concentration of 3,1 mg/mL, 6,25 mg/mL, 12,5 mg/mL, 25 mg/mL, 50 mg/mL, 100 mg/mL 200 mg/mL, 300 mg/mL and S. aureus ATCC@6538TM. This research was conducted using the MIC diffusion method to see the antibacterial activity. UV/Vis spectrophotometry was used to see the antibiofilm activity of butterfly pea extract against S. aureus. The results of the MIC (Minimal Inhibitory Concentration) test showed the antibacterial and antibiofilm activity of butterfly pea extract against S. aureus with a significance value of p=0,001 (p<0,05). The minimum inhibitory concentration of butterfly pea extract is 6,25 mg/mL. The best concentration of butterfly pea extract to inhibit the biofilm formation (antibiofilm) is 100 mg/mL. Butterfly pea extract could cause leakage of DNA, protein, calcium and potassium metals against S. aureus.en_US
