    Pengaruh Mesenchymal Stem Cell terhadap Kadar Gula Darah, Heat Shock Protein 70, Vascular Endothelial Growth Factor, Transforming Growth Factor-β, Endothelial Nitric Oxide Synthase, Jumlah Kolagen, α-Smooth Muscle Actin dan Fungsi Ereksi pada Tikus Wistar Model Diabetic Erectile Dyfunction

    Abstract
    Background Erectile dysfunction (ED) is one of the well-known comorbidities in males with diabetes mellitus which can be induced by dysregulation of corpus cavernosus smooth muscle cells. Umbilical cord mesenchymal stem cells (UCMSCs) are multipotenet cells that attract considerable interest due to immunoregulatory properties and might be a potential strategy to regulate and recover the functional cells and tissues, including tissue improvement in Diabetes mellitus erectile dysfinction (DMED). Objective This study aims to determine the efficacy of UC-MSCs in improving the erectile dysfunction of DMED rats through analyzing the expression of heat shock protein 70, vascular endothelial growth factor, transforming growth factor-β, endothelial nitric oxide synthase, collagen, and α-smooth muscle actin. Methods Total number of 30 male Wistar rats (6 to 8 weeks old) were randomly divided four groups (negative control group, positive control group, T1 group and T2 group). After 16 h fast, 24 rats were randomly selected and intraperitoneally injected with streptozotocin to induced DM. At 8 weeks after STZ injection, rats with DMED were identified by unresponsive erectile stimulation within 30 min. PC group recieved 500 μL; T1 rats treated with 500 μL PBS containing 1X106 UC-MSCs; T2 rats treated with 500 μL PBS containing 3X106 UC-MSCs. After MSCs treatment, the rats were sacrificed and the corpus cavernosum tissues were prepared for histological observations. Results This study resulted in the administration of UC-MSCs could downregulate the regulation leading to the improvement of DMED. Conclusion US-MSCs improve the expression of heat shock protein 70, vascular endothelial growth factor, transforming growth factor-β, endothelial nitric oxide synthase, collagen, and α- smooth muscle actin on erectile dysfunction in streptozocin-induced diabetic rats.
     
    Latar Belakang Disfungsi ereksi (DE) merupakan penyakit yang sering menyertai pria dengan diabetes mellitus yang dapat disebabkan oleh disregulasi sel otot polos korpus kavernosus. Umbilical cord mesenchymal stem cells (UC-MSCs) adalah sel multipoten yang menarik minat yang cukup besar karena sifat imunoregulasi dan mungkin menjadi strategi potensial untuk mengatur dan memulihkan sel dan jaringan fungsional, termasuk perbaikan jaringan pada Diabetes mellitus erectile dysfinction (DMED). Tujuan Penelitian ini bertujuan untuk mengetahui efikasi UC-MSCs dalam memperbaiki disfungsi ereksi tikus DMED melalui analisis ekspresi heat shock protein 70, vascular endothelial growth factor, transforming growth factor-β, endothelial nitric oxide synthase, kolagen, and α-smooth muscle actin. Metode Jumlah tikus Wistar jantan sebanyak 30 ekor (umur 6 sampai 8 minggu) secara acak dibagi empat kelompok (kelompok kontrol negatif, kelompok kontrol positif, kelompok T1 dan kelompok T2). Setelah puasa 16 jam, 24 tikus dipilih secara acak dan disuntik secara intraperitoneal dengan streptozotocin untuk menginduksi DM. Pada 8 minggu setelah injeksi STZ, tikus dengan DMED diidentifikasi oleh stimulasi ereksi yang tidak responsif dalam waktu 30 menit. Kelompok PC menerima 500 μL; Tikus T1 yang diberi 500 μL PBS yang mengandung 1X106 UC-MSCs; Tikus T2 diobati dengan 500 μL PBS yang mengandung 3X106 UC-MSCs. Setelah pengobatan MSC, tikus dikorbankan dan jaringan corpus cavernosum disiapkan untuk pengamatan histologis. Hasil Penelitian ini menemukan UC-MSCs dapat meregulasi perbaikan DMED. Kesimpulan US-MSCs meningkatkan ekspresi heat shock protein 70, vascular endothelial growth factor, transforming growth factor-β, endothelial nitric oxide synthase, kolagen, dan α-smooth muscle actin pada disfungsi ereksi pada tikus diabetes yang diinduksi streptozocin.
