    Uji Efek Imunomodulator Kombinasi Ekstrak Ikan Gabus (Channa striata), Meniran (Phyllanthus niruri), dan Temulawak (Curcuma xanthorrhiza) terhadap Aktivitas Fagositosis Sel Imun pada Mencit Jantan dengan Metode Bersihan Karbon (Carbon Clearance)

    Abstract
    Background: Immunomodulators are compounds that affect the work of the immune system by stimulating the immune system. The content of secondary metabolites such as flavonoids from a combination of extract of snakehead fish (Channa striata), meniran (Phyllanthus niruri), and temulawak (Curcuma xanthorrhiza) act as immunostimulants in the order they can increase metabolic activity in cells. Objective: To determine the effect of the combination of snakehead fish extract (Channa striata), meniran (Phyllanthus niruri), and temulawak (Curcuma xanthorrhiza) on increasing phagocytic activity in male mice. Methods: The immunomodulatory effect was tested using the carbon clearance method. They were divided into 5 treatment groups, namely CMC Na 0.5%, Imboost 32.5 mg/kg BW, and a combination of extract doses of 130, 390, and 780 mg/kg BW. Mice were given oral treatment for 7 days. On the eighth day, carbon suspension was given IV. After that, at the 5th minute, blood was drawn, followed by 10, 15, and 20 minutes. Then the absorbance was measured using a UV-Vis spectrophotometer, the mice were dislocated and dissected, and then the liver and liver were taken. spleen. The rate of carbon elimination, phagocytosis index, and stimulation index were calculated. Results: The results showed that the administration of a combination of EIGMT doses of 130, 390, and 780 mg/kg BW could increase the rate of carbon elimination, phagocytosis index, and stimulation index when compared to the negative control CMC Na 0.5%, which was significantly different (p < 0,05). The combination of EIGMT at a dose of 780 mg/kg BW had the highest rate of elimination compared to a combination of EIGMT at a dose of 130 and 390 mg/kg BW. The phagocytosis index of the combined EIGMT doses of 130, 390, and 780 mg/kg BW was 3.35, respectively; 4.12 ; 4.61 mg/kg BW. The combination stimulation index of EIGMT doses of 130, 390, and 780 mg/kg BW, respectively, was 1.18; 1.45 ; and 1.62. Conclusion: The combination of extracts has an effect on increasing phagocytic activity in male mice. The combination of EIGMT doses of 130, 390, and 780 mg/kg BW can increase phagocytic activity. The combination of an EIGMT dose of 780 mg/kg BW gave almost the same effect as the positive control Imboost 32.5 mg/kg BW.
     
    Latar Belakang: Imunomodulator adalah senyawa yang mempengaruhi kerja sistem kekebalan tubuh dengan meningkatkan sistem imun. Kandungan metabolit sekunder seperti flavonoid dari kombinasi ekstrak ikan gabus (Channa striata), meniran (Phyllanthus niruri), dan temulawak (Curcuma xanthorrhiza) berperan sebagai imunostimulan sehingga dapat meningkatkan aktivitas metabolisme di dalam sel. Tujuan: Untuk mengetahui pengaruh kombinasi ekstrak ikan gabus (Channa striata), meniran (Phyllanthus niruri), dan temulawak (Curcuma xanthorrhiza) dapat meningkatkan aktivitas fagositosis pada mencit jantan. Metode: Pengujian efek imunomodulator dilakukan dengan metode bersihan karbon (Carbon Clearance) di bagi menjadi 5 kelompok perlakuan, yaitu CMC Na 0.5%, Imboost 32,5 mg/kg BB, dan kombinasi ekstrak dosis 130, 390, dan 780 mg/kg BB. Mencit diberikan perlakuan oral selama 7 hari. Pada hari ke delapan diberikan suspensi karbon secara i.v, setelah itu pada menit ke 5 dilakukan pengambilan darah, dilanjutkan pada menit ke-10, 15, dan 20. Kemudian absorbansinya diukur menggunakan spektrofotometer UV-Vis, kemudian mencit didislokasi dan dibedah lalu diambil hati dan limfa. Dihitung laju eliminasi karbon, indeks fagositosis, dan indeks stimulasi. Hasil: Hasil penelitian menunjukkan bahwa pemberian kombinasi EIGMT dosis 130, 390, dan 780 mg/kg BB dapat meningkatkan laju eliminasi karbon, indeks fagositosis dan indeks stimulasi bila dibandingkan terhadap kontrol negatif CMC Na 0,5% yang berbeda secara signifikan (p < 0,05). Kombinasi EIGMT dosis 780 mg/kg BB memiliki laju eliminasi yang paling tinggi dibandingkan dengan kombinasi EIGMT dosis 130 dan 390 mg/kg BB. Indeks fagositosis kombinasi EIGMT dosis 130, 390 dan 780 mg/kg BB berturut-turut adalah 3,35 ; 4,12 ; 4,61 mg/kg BB. Indeks stimulasi kombinasi EIGMT dosis 130, 390 dan 780 mg/kg BB secara berurutan adalah 1,18 ; 1,45 ; dan 1,62. Kesimpulan: Kombinasi ekstrak mempunyai pengaruh terhadap peningkatan aktivitas fagositosis pada mencit jantan. Kombinasi EIGMT dosis 130, 390 dan 780 mg/kg BB dapat meningkatkan aktivitas fagositosis. Kombinasi EIGMT dosis 780 mg/kg BB memberikan efek yang hampir sama dengan kontrol positif Imboost 32,5 mg/kg BB.
