Formulasi Sediaan Gel Ekstrak Etanol, Fraksi N- Heksan dan Fraksi Etil Asetat Biji Jintan Hitam (Nigella Sativa L.) Sebagai Tonik Rambut

Abstract

Hair Tonic is a cosmetic preparation to help strengthen and improve hair growth. Black cumin (Nigella sativa L.) contains oleic and linoleic acids which can inhibit hair loss. Hair tonics in gel dosage forms are in demand because of their effective, practical and convenient use when applied. The purpose of this study was the formulation of ethanol extract gel, n-hexane fraction and ethyl acetate fraction of black cumin seeds as hair tonic. The black cumin seeds were dried then pollinated to become simplicia, then characterized and macerated with 80% ethanol. The extract was concentrated using a rotary evaporator, then fractioned with n-hexane and ethyl acetate solvents. This test used the concentration of ethanol extract, n-hexane and ethyl acetate fraction, respectively 3%, 5%, 7% and used positive control and negative control, each formula was applied to 5 rats. Before being applied, the rats' hair was shaved first, then the gel evaluation test included organoleptic, homogeneity, pH, viscosity and dispersibility tests. availability evaluation includes irritation test, waterproof effectiveness test, microbial contamination test and hair growth test. The parameters measured were hair length, hair weight, hair thickness and the number of hairs in the pores. The treatment is carried out for 21 days by applying the preparation once a day. The results of the simplicia characterization test meet the requirements of the Indonesian Natural Cosmetics Material. Hair tonic gel preparation is stable in storage for 12 weeks and meet the requirements of SNI 16-4955-1998. The hair growth activity in the gel preparation with n-hexane fraction of 7% against positive control showed no significant difference (p> 0.05), with hair length of 2.11 ± 0.01mm and hair weight of 0.0780 ± 0.008mg. The conclusion of this study is the hair tonic gel preparation of ethanol extract, n-hexane fraction and ethyl acetate fraction fulfilled SNI 16-4955-1998, and the n-hexane fraction with a concentration of 7% had more effective hair growth activity than other concentrations.

Tonik Rambut merupakan sediaan kosmetik untuk membantu menguatkan, memperbaiki pertumbuhan rambut. Jintan hitam (Nigella sativa L.) mengandung asam oleat dan linoleat yang dapat menghambat kerontokan rambut. Tonik rambut dalam bentuk sediaan gel diminati karena penggunaannya yang efektif, praktis dan nyaman saat dioleskan. Tujuan penelitian ini adalah formulasi sediaan gel ekstrak etanol, fraksi n-heksan dan fraksi etil asetat biji jintan hitam sebagai tonik rambut. Biji jintan hitam dikeringkan kemudian diserbukkan hingga menjadi simplisia, kemudian dikarakterisasi dan dimaserasi dengan etanol 80%. Ekstrak di pekatkan dengan rotary evaporator, kemudian di fraksi dengan pelarut n-heksan dan etil asetat. Pengujian ini menggunakan konsentrasi sediaan ektrak etanol, fraksi n-heksan dan fraksi etil asetat masing-masing 3%, 5%, 7% serta menggunakan kontrol positif dan kontol negatif, tiap formula diaplikasikan pada 5 ekor tikus. Sebelum dioleskan rambut tikus dicukur terlebih dahulu, kemudian di uji evaluasi gel meliputi uji organoleptis, homogenitas, pH, viskositas dan daya sebar. Evaluasi sediaaan meliputi uji iritasi, uji efektivitas tahan air, uji cemaran mikroba dan uji pertumbuhan rambut. Parameter yang diukur yaitu panjang rambut, bobot rambut, ketebalan rambut dan jumlah rambut dalam pori. Perawatan dilakukan selama 21 hari dengan mengaplikasikan sediaan satu kali sehari . Hasil uji karakterisasi simplisia memenuhi persyaratan Materia Kosmetika Bahan Alam Indonesia. Sediaan gel tonik rambut stabil dalam penyimpanan selama 12 minggu dan memenuhi syarat SNI 16-4955-1998. Aktivitas pertumbuhan rambut pada sediaan gel fraksi n-heksan 7% terhadap kontrol positif menunjukkan tidak adanya perbedaan yang signifikan (p>0,05), dengan panjang rambut 2,11±0,01mm dan bobot rambut 0,0780±0,008mg. Kesimpulan dari penelitian ini adalah sediaan gel tonik rambut ekstrak etanol, fraksi n-heksan dan fraksi etil asetat memenuhi SNI 16-4955-1998, dan fraksi n-heksan dengan konsentrasi 7% memiliki aktivitas pertumbuhan rambut yang lebih efektif dari konsentrasi lainnya.
