Efek Kombinasi Gel Ekstrak Etanol Daun Binahong (Anredera cordifolia) dan Daun Mobe (Artocarpus lakoocha) dengan Perbandingan 1:1 terhadap Proliferasi Fibroblas dan Osteosit pada Penyembuhan Luka Soket Pasca Pencabutan Gigi Tikus Wistar

Abstract

Herbal plants that can be used for wound healing are binahong leaves (Anredera cordifolia) and mobe leaves (Artocarpus lakoocha). Both of these leaves contain secondary metabolites that can help the wound healing process. However, there has been no research that has studied the effect of the combination of binahong leaves and mobe leaves on the healing of tooth socket wounds. The purpose of this study was to determine the effect of the combination of ethanol extract gel from binahong leaves (Anredera cordifolia) and mobe leaves (Artocarpus lakoocha) in a 1:1 ratio on the proliferation of fibroblasts and osteocytes in socket wounds after tooth extraction. This study is a laboratory experimental study with a post test only control group research design. The samples used were 16 male Wistar rats (Rattus norvegicus) that had been acclimatized. Tooth extraction of male Wistar rats was carried out on the lower left incisor, then gel was applied to the four observation groups with two different treatments, a combination of binahong and mobe leaf extract gel leaf and Aloclair® gel (control). Each group was observed with Hematoxylin Eosin examination to see the proliferation of fibroblasts and osteocytes on the 7th and 21st days after tooth extraction of rats. The research data were analyzed using independent T-test. The results showed that there were significant difference in the average number of fibroblasts and osteocytes in the treatment group with the combination of binahong leaf extract and mobe leaf extract gel and the control group with Aloclair® gel (p<0.05). The conclusion of this study was that the combination of ethanol extract gel of binahong leaves (Anredera cordifolia) and mobe leaves (Artocarpus lakoocha) with a ratio of 1:1 was able to increase the proliferation of fibroblasts and osteocytes in the healing of socket wounds after tooth extraction.

Tanaman herbal yang dapat digunakan pada penyembuhan luka yaitu daun binahong (Anredera cordifolia) dan daun mobe (Artocarpus lakoocha). Kedua daun ini memiliki kandungan metabolit sekunder yang dapat membantu proses penyembuhan luka. Namun, belum ada penelitian yang mempelajari tentang pengaruh kombinasi daun binahong dan daun mobe terhadap penyembuhan luka soket gigi. Tujuan dari penelitian ini adalah untuk mengetahui efek pemberian kombinasi gel ekstrak etanol daun binahong dan daun mobe dengan perbandingan 1:1 terhadap proliferasi fibroblas dan osteosit pada luka soket pasca pencabutan gigi tikus. Penelitian ini merupakan penelitian eksperimental laboratorium dengan desain penelitian post test only control group. Sampel yang digunakan sebanyak 16 ekor tikus Wistar jantan yang sudah diaklimatisasi. Pencabutan gigi tikus Wistar jantan dilakukan di insisivus kiri rahang bawah, kemudian diaplikasikan gel pada empat kelompok pengamatan dengan dua perlakuan berbeda, kombinasi gel ekstrak daun binahong dan daun mobe dan gel Aloclair® (kontrol). Masing-masing kelompok diamati dengan pemeriksaan Hematoxylin Eosin untuk melihat proliferasi sel fibroblas dan osteosit pada hari ke-7 dan hari ke-21 pasca pencabutan gigi tikus. Data penelitian dianalisis menggunakan uji-T tidak berpasangan. Hasil penelitian menunjukkan terdapat perbedaan rata-rata jumlah sel fibroblas dan sel osteosit yang signifikan pada kelompok perlakuan dengan kombinasi gel ekstrak daun binahong dan daun mobe dan kelompok kontrol dengan gel Aloclair (p<0,05). Kesimpulan penelitian ini adalah kombinasi gel ekstrak etanol daun binahong (Anredera cordifolia) dan daun mobe (Artocarpus lakoocha) dengan perbandingan 1:1 mampu meningkatkan proliferasi fibroblas dan osteosit pada penyembuhan luka soket pasca pencabutan gigi.
