The dried crude material (root, stem and leaf) of sambiloto (Andrographis paniculata Nees) have a positive cholekinetic effect. The aim of the present study was to compare the cholekinetic effect of herba dried crude material and ethanol extract of sambiloto. A randomized, double blind and crossover design study was carried out in 10 healthy volunteers. After an overnight fast (± 10 hours), the gallbladder volume (GBV) were ultrasonographically measured at 08.00 AM and then at 0.5, 1.0, 1.5 and 2.0 hour after administration of 250 mg of herba dried crude material, 10 mg, and 20 mg of extract. Data obtained were expressed as mean ± standard deviation and statistically analysed by one-way ANOV A, with p-values less than 0.05 regarded as a statistically significant. The fasting GBV before taking 250 mg of dried crude material, 10 mg and 20 mg of extract and placebo were 9.18 ± 3.33 ml, 7.79 ± 2.46 ml, 10.36 ± 2.19 ml, and 10.32 ± 2.09 ml respectively, which not statistically different (p = 0.101). After administration of sambiloto, GBV were decrease by increasing observation time, but at placebo group the GBV were increase after 2 hour of administration. The percentage of gallbladder contraction (GBC) after 0.5 hour of administration of 250 mg of herba dried crude material (21.69 ± 19.02 %), 10 mg (29.08 ± 30.35 %) and 20 mg of extract (37.62 ± 12.57%) (p< 0.01). After 2 hour, the percentage of GBC in groups that received 20 mg of extract (75.49 ± 7.96%) which higher than 250 mg of herba dried crude material (59.45 ± 15.09%) and 10 mg of extract (66.67 ± 13.61%). The present study clearly demonstrated that herba dried crude material and ethanol extract of sambiloto have positive cholekinetic effect. The cholekinetic effect of extract were higher than herba dried crude material.
