The increase in potato production in Indonesia is constrained by the conditions for growing potatoes which are only met in areas with an altitude of 1500 meters above sea level. It is known that at high temperatures the GA content in plants will increase thereby inhibiting tuber formation. The use of growth inhibitors such as paclobutrazol was able to inhibit GA biosynthesis so as to stimulate tuber formation. This study aims to determine the effect of paclobutrazol on the induction of potato microtubers (Solanum tuberosum L) at various incubation temperatures. This research was conducted at the Laboratory of Tissue Culture and Greenhouse, Faculty of Agriculture, North Sumatra. This study used a randomized block design with two factors. The first factor was the administration of Paclobutrazol with five levels, namely: 0 ppm, 7.5 ppm, 15 ppm, 22.5 ppm, 30 ppm. The second factor is incubation temperature with three levels, namely: laboratory temperature (18,25–18,75oC), room temperature (22,50–27,00oC), and greenhouse temperature (29,50–33,50oC). Parameters observed were tuber emergence time, plant height, number of branches, number of tubers, tuber diameter and tuber weight. The results showed that the administration of paclobutrazol at a dose of 7.5 ppm showed a higher average than the other doses in the number of tubers. The incubation temperature at laboratory temperature showed the highest results compared to room temperature and greenhouse temperature at the time of emergence of tubers and tuber diameter.
