This study aims to obtain information about the genetic diversity of durian plants in several districts in North Sumatra using RAPD markers. This research was conducted at the Laboratory of Molecular Genetics, Faculty of Agriculture, University of North Sumatra, Medan from February to April 2022. The plant samples came from 6 districts in North Sumatra, namely Deli Serdang, Langkat, Central Tapanuli, Dairi, South Tapanuli and Humbang Hasundutan regencies. Determination of band size using UVITEC Cambridge Fire Reader software and determination of genetic similarity and group formation using Multivariate Statistical Package (MVSP ver 3.2) software. Based on 12 samples of durian plants isolated and amplified using primers OPB 18, OPN 14 and OPD 20. Primer OPB 18 obtained a band size of 300-750 bp, primer OPN 14 obtained a band size of 400-600 bp, and primer OPD 20 obtained a band size of 550 -700 bp. Based on the phylogenetic results with UPGMA, all samples analyzed were divided into 2 groups. Group (I) consists of SHH. Group
(II) consists of BTS, PTS, SD, PHH, BD, STT, PTT, SDS, KL, JL, BDS. The results of the analysis showed that 12 samples with genetic similarity values ranged from 0.54-1
