    Isolasi dan Formulasi Sediaan Hidrogel menggunakan Kitosan dari Tulang Sotong (Sepia Sp.) terhadap Luka Sayat Eksisi Tikus Putih Jantan

    Abstract
    Hydrogels have elastic, hydrophilic properties and are able to expand and absorb large amounts of water so that they can be used as good conductors of active ingredients such as chitosan. Cuttlefish bone is one source of natural ingredients that contain chitosan. Chitosan is able to act as a wound healer such as excision cuts. The purpose of this study was to evaluate the effectiveness of cuttlefish bone chitosan hydrogel on excision wound healing in male white rats. Chitosan was isolated using acid and alkaline solvent from cuttlefish bone, then characterized by water content, ash content, protein content, ninhydrin test and functional groups. The chitosan obtained was made into hydrogel preparations with a concentration of 0.5% (F1), 1% (F2), 1.5% (F3). Evaluation of hydrogel preparations included organoleptic, pH, physical characteristics, cross linking, swelling, elasticity, Cycling Test. The effectiveness of the excision wound was tested on male white rats and measured the diameter of the wound and the percentage of the excision wound. The results of the characterization of chitosan have a water content value of 8.04%, ash content of 1.73% and protein content of 4.8%. The best evaluation of hydrogel preparations was found in cuttlefish bone chitosan concentration of 0.5% (F1), did not change color, during storage with an average pH value of 4.52. The results of the characterization of 0.5% chitosan hydrogel (F1) were 8.5 cm in diameter, 0.45 mm in thickness and >200 times the folding power. 0.5% chitosan hydrogel (F1) showed 1.3% cross-link, 74.52% swelling, 14.2% elasticity. Cuttlefish bone chitosan hydrogel 0.5% (F1) can heal rat excision wounds by increasing the percentage reduction in wound diameter and reduction in wound diameter. The administration of 0.5% cuttlefish bone chitosan hydrogel gave a significant difference to the positive control and negative control (p>0.05%). The conclusion of this study is that 0.5% cuttlefish bone chitosan hydrogel is the best formula and has the effectiveness of heal excision wounds in male white rats seen from the reduction in wound diameter and an increase in the percentage reduction in wound diameter.
