dc.description.abstractPapaya (Carica papaya L) male flower, is one of the medicinal plants in North Sumatra used to treat degenerative and metabolic diseases. Previous research has been done to test the potential of ethanol extracts on the inhibition of tumor growth or breast cancer in Sprague-Dawley female rats. This study aims to determine the activity that triggerapoptosis of ethanol extract, fraction of n-hexane and ethyl acetate fraction of papaya male flower in colon cancer cells (WiDr) through cytotoxic test, selectivity index, apoptosis triggering and suppression of expression of cyclin D1 and Bcl-2 with doxorubicin as the comparison. Preparation of the extract conducted by percolation with 80% ethanol and then fractionated using a solvent based on the degree of polarity, n-hexane, ethyl acetate and the characterization of the crude drugwas also conducted. The citotoxic of the extract against WiDr cells and Vero cells was tested using MTT (Microculture Tetrazolium Technique), colorimetric method, while selectivity index and apoptosis were tested by using flow cytometry methods, and suppression of expression of cyclin D1 and Bcl-2 by immunocytochemistry method. The test result of FHBPJ cytotoxic in colon cancer cells of WiDr provided IC50 value of 64,105 μg/ml. Cytotoxic test against normal cells (Vero cells) showed FHBPJ was selective against Vero cells (IS> 3), and coudtrigger apoptosis with the percentage of 93,55%; 91,81%; 2,67% and 1,47% and could suppress the expression of cyclin D1 and Bcl-2. Based on these results, so FHBPJ can be used as colon anticancer for their synergistic effect and the triggering of apoptosisen_US
dc.description.abstractBunga pepaya jantan (Carica papaya L), merupakan salah satu tumbuhan obat di Sumatera Utara yang digunakan untuk mengatasi penyakit degeneratif dan metabolisme. Penelitian sebelumnya telah melakukan uji potensi eksrak etanol terhadap penghambatan pertumbuhan tumor atau kanker payudara pada tikus betina Sprague Dawley. Penelitian ini bertujuan untuk mengetahui aktivitas pemicu apoptosis ekstrak etanol, fraksi n-heksan dan fraksi etil asetat bunga pepaya jantan pada sel kanker kolon (WiDr) melalui uji sitotoksik, indeks selektivitas, pemacuan apoptosis dan penekanan ekspresi siklin D1 dan Bcl-2 dengan pembanding doksorubisin. Pembuatan ekstrak dilakukan dengan cara perkolasi dengan pelarut etanol 80% dan kemudian difraksinasi menggunakan pelarut berdasarkan tingkat kepolarannya, n-heksana, etil asetat serta dilakukan karakterisasi terhadap simplisia. Fraksi n-Heksan diuji sitotoksiknya terhadap sel WiDr dan sel VeRo dengan menggunakan metode kolorimetri MTT (Microculture Tetrazolium Technique), indeks selektivitas, dan apoptosis dengan metode flowsitometri serta penekanan ekspresi siklin D1 dan Bcl-2 dengan metode imunositokimia. Hasil uji sitotoksik FHBPJ pada sel kanker kolon WiDr memberikan nilai IC50 sebesar 64.105 μg/ml. Pengujian sitotoksik terhadap sel normal (sel Vero) menunjukkan FHBPJ selektif terhadap sel VeRo (IS > 3), serta dapat memacu apoptosis dengan persentase 93,55%; 91,81%; 2,67% dan 1,47% dan dapat menekan ekspresi siklin D1 dan Bcl-2. Berdasarkan hasil tersebut maka FHBPJ dapat sebagai antikanker kolon karena adanya efek sinergis dan terjadinya pemacuan apoptosis.en_US
