Efek Relaksasi Ekstrak Dan Fraksi Daun Pugun Tanoh (Picria fel-terrae Lour.) Pada Otot Polos Ileum Tikus Terisolasi Secara In Vitro Yang Dikontraksikan Dengan Serotonin

Abstract

Pugun Tanoh (Picria fel-terrae Lour.) is a plant of Linderniaceae family which is often used by the native people of Tiga Lingga village in Dairi Regency, North Sumatera Province, Indonesia to cure diarrhea. This research aims to observe the relaxation effect of extract and fraction of pugun tanoh leaves and its effect on PGE2 and NO on the contraction of isolated rat’s ileum smooth muscle contracted by serotonin. This research was conducted using in vitro, the parameter measured in this research is the contraction or relaxation of isolated rat’s ileum smooth muscle. The relaxation effect test is done after the rat’s ileum is contracted with serotonin, then each of the ileum is given the Pugun Tanoh leaf ethanol extract (EEDPT), n-hexane fraction (FnHDPT), ethyl acetate fraction (FEADPT), and residu fraction cumulative concentrate. The effect of PGE2 and NO was done by incubating the ileum with aspirin and L-NNA, then ileum was contracted with serotonin, and given the cumulative concentration of the active fraction. The result of the research shows that EEDPT, FnHDPT, FEADPT, and FSDPT at 0,5 - 4 mg/ml concentration has relaxation effect, where relaxation effect of FEADPT better than other extracts, and FEADPT at 2,5 mg/ml concentration has no difference in terms of ability as ondansetron 3 x 10-2 mg/ml in reducing the ileum smooth mucle contraction induced by serotonin 1,08 x 10-6 M. The relaxation effect of FEADPT on ileum smooth muscle contracted with serotonin is not affected by PGE2, but affected by NO.

Pugun tanoh (Picria fel-terrae Lour.) merupakan tanaman dari suku Linderniaceae yang sering digunakan oleh masyarakat Desa Tiga Lingga, Kabupaten Dairi, Provinsi Sumatera Utara, Indonesia untuk mengobati diare. Penelitian ini bertujuan untuk mengetahui efek relaksasi dari ekstrak dan fraksi daun pugun tanoh, serta pengaruhnya terhadap PGE2 dan NO pada kontraksi ileum tikus terisolasi yang dikontraksikan dengan serotonin. Penelitian dilakukan secara in vitro, parameter yang diukur dalam penelitian ini adalah kontraksi atau relaksasi otot polos ileum tikus terisolasi. Pengujian efek relaksasi dilakukan setelah ileum dikontraksikan dengan serotonin, kemudian masing – masing ileum diberikan konsentrasi kumulatif ektrak etanol (EEDPT), fraksi n-heksan (FnHDPT), fraksi etil asetat (FEADPT), dan fraksi sisa (FSDPT) daun pugun tanoh. Uji pengaruh PGE2 dan NO dilakukan dengan menginkubasi ileum dengan aspirin dan L-NNA, kemudian ileum dikontraksikan dengan serotonin, dan diberikan konsentrasi kumulatif fraksi aktif. Hasil penelitian EEDPT, FnHDPT, FEADPT, dan FSDPT konsentrasi 0,5 - 4 mg/ml memiliki efek relaksasi, dimana FEADPT merelaksasi otot polos lebih baik dibanding ketiga ekstrak lainnya, dan FEADPT pada konsentrasi 2,5 mg/ml tidak berbeda dengan ondansetron pada konsentrasi 3 x 10-2 mg/ml untuk menurunkan kontraksi otot polos yang dikontraksikan dengan serotonin 1,08 x 10-6 M (p>0,05). Efek relaksasi FEADPT pada otot polos ileum yang dikontraksikan dengan serotonin tidak dipengaruhi PGE2, tetapi dipengaruhi NO.
