Isolasi dan Identifikasi Bakteri Kitinolitik dari Nepenthes tobaica dan Nepenthes gracilis

Nepenthes, a pitcher plant obtains its nitrogen from degrading trapped insect in the
pitcher. Chitinolytic bacteria are assumed acting in degradation of exoskeletons of
insects in the pitcher. In this study, chitinolytic bacteria were isolated from the
fluid of opened and closed pitchers, pitchers, stalks, roots, and rhizosphere soil of
Nepenthes tobaica and Nepenthes gracilis from Merek, Dairi, North Sumatra.
Total of 18 isolates were obtained by isolating the bacteria on minimum salt
media chitin adding with 0.2% yeast extract. Two isolates that have higher
chitinolytic index, AM1 was from the root of N. tobaica with chitinolytic index of
2.25, and CbM was from the fluid of opened pitcher of N. tobaica with
chitinolytic index of 1.90. Identification of 16S rRNA gene indicated that CbM
isolate was closely related to Enterobacter aerogenes strain VT66, while AM1
was not succeeded to be identified.
