Cellulase is a component consisting of a complex mixture of enzymes responsible for hydrolyze a cellulose. Cellulase enzymes was produced from hydrolysis of cellulose which will affect bioethanol yield during fermentation. Mixed culture of Trichoderma reesei and Aspergillus niger performed the best cellulase activities. The study aim to learn the effect of incubation time during hydrolysis of corn cobs into glucose by using Trichoderma reesei, Aspergillus niger, and a mixture of both. This study used corn cobs as a feedstock. Corn cobs were autoclaved for 15 minutes at the temperature of 120 oC. Trichoderma reesei and Aspergillus niger were cultured in PDA medium for 72 hours at the temperature of 20 oC. Inoculum were incubated for 48 hours at the temperature of 20 oC. The affect of the ratio of the volume of microbes and incubation time were observed in this study. The best enzyme activity obtained from combination of Trichoderma reesei and Aspergillus niger is 2:1 with 5 days incubation time.en_US
